复合异种骨移植的实验研究

摘    要:目的探讨醋酸甲基泼尼松龙(Methyl―Prednisolone Acetate MPA)与部分脱钙异种骨复合在骨缺损修复中诱导成骨及免疫排斥反应进行本项研究.方法将40只成年新西兰兔随机分成两大组.各大组再随机分成五小组,手术造成兔双侧桡骨中段8mm骨缺损.其中一大组动物右侧骨缺损植入复合异种骨(部分脱钙猪骨加MPA),另一大组动物右侧骨缺损植入单纯异种骨(部分脱钙猪骨);两组左侧均植入自体新鲜骨,两大组动物分别于术后1、2、4、6、8周处死,标本作病理切片观察植骨周围组织学变化及形态学变化,两大组8周末处死的8只动物于术前及术后2、4、6周各抽心脏血一次,行IL-2活性及T细胞转化测定,结果 X线摄片复合异种骨侧第二周末骨缺损区可见密度增高影,第四、六周末可见骨钙化影充填并连接断端,第八周末部分动物骨缺损区骨髓腔再通,与自体新鲜骨侧变化类似.结论复合异种骨侧各时间植骨周围淋巴细胞及浆细胞浸润较单纯异种骨组明显少,IL-2活性测定,T淋巴细胞转化统计学结果示,复合异种骨与单纯异种骨组间有显著性差异,说明醋酸甲基泼尼松龙具有诱导成骨作用又在局部应用抑制免疫排斥反应.

Abstract:Objective To observe the immunity rejection and osteogeneic repair of a composite bone graft in bone defects. Methods Forty adult New Zealand rabbits were divided into two groups, the composite bone was implanted in the right radii defects, Partially decalcified xenogeneic bone was similarly implanted in the right defects. In the other group, in all rabbits, autologous fresh bone was implanted in the left, Radii defects. Results At the end of the 1st, 2nd 4th, 6th and 8th week after Operation. We observe the tissue around the implanted bone; the rabbits killed at 8th week were drawn heart blood at preoperative, 2nd, 4th and 6th week to measure IL-2 active and T cell transformation. Conclusion Methyl- Prednisolone Acetate applied in location graft have been proven to restrain immunity rejection and Stimulate Osteogeneic repair.
