过表达长链非编码RNA H19对肝癌细胞增殖和自噬的影响及可能机制

摘    要:目的:研究长链非编码RNA H19(long noncoding RNA H19,H19)对肝癌细胞自噬的影响及可能机制。方法:Real-time PCR检测H19在肝癌组织及相应癌旁组织中的表达;CCK-8分析过表达H19对肝癌细胞增殖的影响;免疫荧光实验和Western blot分析过表达H19对肝癌细胞自噬的影响;Western blot分析过表达H19肝癌细胞的Atg7表达变化以及与Atg7干扰质粒共转染后对肝癌细胞自噬的影响。结果:H19在肝癌组织中表达水平高于癌旁组织;过表达H19后肝癌细胞增殖活力及自噬增强;过表达H19通过上调自噬相关基因Atg7促进肝癌细胞自噬。结论:上调H19促进肝癌细胞增殖与自噬,H19对肝癌细胞自噬的调控可能与Atg7有关。

Abstract:Objective:To explore the regulation of long noncoding RNA H19 (H19) on cell proliferation and autophagy of hepatocellular carcinoma.Methods:Real-time PCR was used to measure the expression of H19 in hepatocellular carcinoma tissues and adjacent tissues.The proliferation level of hepatocellular carcinoma cells was analyzed by CCK-8.Immunofluorescence assay and Western blot were used to analyze autophagy activity.Western blot was used to analyze the expression of Atg7 and the effect of co-transfection with Atg7 on autophagy of hepatocellular carcinoma cells.Results:The expression level of H19 was up-regulated in hepatocellular carcinoma tissues than in the adjacent tissues.Overexpressed H19 could promote cells proliferation and autophagy of hepatocellular carcinoma cells.The expression of Atg7 could be enhanced by overexpressed H19.Conclusion:H19 could promote proliferation and autophagy in HCC cells.The promotion of H19 on autophagy might be related to Atg7.
