Fzd2诱导ER阴性乳腺癌细胞上皮-间质转化

目的:分析Frizzled 2（Fzd2）在雌激素受体阴性（ER-）乳腺癌中的表达及其对ER-乳腺癌细胞上皮-间质转换（epithelial-mesenchymal transition，EMT）的影响。方法:采用免疫组化方法检测Fzd2在乳腺癌组织中的表达；采用Western blot检测Fzd2、E-cadherin、Vimentin、Slug、Zeb1在乳腺癌细胞中的表达；采用伤口愈合试验和Transwell试验检测细胞迁移和侵袭。结果:Fzd2在ER-乳腺癌组织及细胞系中过表达；Fzd2敲减促进Hs578T细胞表达E-cadherin，减少Hs578T细胞表达Vimentin、Slug和Zeb1；Fzd2敲减抑制Hs578T细胞迁移和侵袭。结论:Fzd2诱导ER-乳腺癌细胞发生EMT；Fzd2可能作为ER-乳腺癌防治的一个潜在靶向分子。

Fzd2 induces epithelial-mesenchymal transition in ER-negative breast cancer

Objective:To investigate the expression of Frizzled 2 (Fzd2) in ER-negative (ER-) breast cancer,and to explore the effect of Fzd2 on the epithelial-mesenchymal transition (EMT) in ER- breast cancer cells.Methods:Immunohistochemistry was used to detect the expression of Fzd2 in ER- and ER+ breast cancer tissues.Western blot was used to examine the expression of Fzd2,E-cadherin,Vimentin,Slug and Zeb1 in breast cancer cells.Wound healing test and Transwell test were performed to analyze breast cancer cell migration and invasion.Results:Fzd2 was overexpressed in ER- breast cancer tissues and cells.Fzd2 knockdown enhanced the expression of E-cadherin,while reduced the expression of Vimentin,Slug and Zeb1 in Hs578T cells.Fzd2 knockdown suppressed migration and invasion of Hs578T cells.Conclusion:Fzd2 induces EMT in ER- breast cancer cells,and Fzd2 may be a potential target for prevention and therapy of ER- breast cancer.
