WBP5在胃癌组织中的表达与临床病理特征的相关性及其对胃癌细胞迁移侵袭的影响

目的:探讨WBP5在胃癌组织中的表达与临床病理特征的相关性及其对胃癌细胞迁移、侵袭等的影响。方法:通过免疫组化检测WBP5在胃癌癌组织及癌旁组织中的表达，分析WBP5与胃癌临床病理特征的关系。通过细胞转染技术，建立WBP5稳定高表达和低表达的细胞，分析WBP5对癌细胞划痕愈合、迁移及侵袭能力的影响，及其对EMT相关指标的影响。结果:WBP5在癌组织中呈低表达，在癌旁组织中高表达，WBP5的表达和胃癌患者的T分期及TNM分期显著相关。上调WBP5后癌细胞划痕愈合比例显著降低，迁移、侵袭能力显著降低；下调WBP5后癌细胞划痕愈合比例显著上升，迁移、侵袭能力显著升高。上调WBP5后E-cadherin表达上调，N-cadherin及Vimentin表达下调；下调WBP5后E-cadherin表达下调，N-cadherin及Vimentin表达上调。结论:WBP5表达与患者的临床分期显著相关，WBP5表达与胃癌细胞的迁移、侵袭能力有重要关系，其机制可能是通过影响EMT过程实现的，具体机制需要进一步研究。

Expression of WBP5 in gastric cancer tissues and the correlation with clinicopathological characteristic and the effect on the migration and invasion of gastric cancer cells

Objective:To detect the expression of WBP5 in gastric cancer tissues and the correlation with clinicopathological characteristic，investigate its effects on migration and invasion of gastric cancer cells．Methods:Immunohistochemical technique was conducted to investigate the clinical features of WBP5 expression in human gastric cancer tissues.Stable WBP5-overexpressed and WBP5-downexpressed cells were established to assess the role of WBP5 on the migration and invasion and the effect of EMT of gastric cancer cells.Results:WBP5 expression was lower in cancer tissue than para-cancer tissue.WBP5 was significantly associated with T staging and TNM staging of gastric cancer patients.Up-regulation WBP5 decreased the healing rate and the migration and invasion of gastric cancer cell,while down-regulation WBP5 enhanced the healing rate and the migration and invasion of gastric cancer cell.Up-regulation WBP5 can lower the expression of N-cadherin and Vimentin and increase the expression of E-cadherin.Down-regulation WBP5 can increase the expression of N-cadherin and Vimentin and lower the expression of E-cadherin.Conclusion:WBP5 expression is correlated with clinical stage in gastric cancer patients.WBP5 could modulate the migration and invasion of gastric cancer cells and the mechanism may be through EMT.The mechanism of WBP5 need further explore.
