STAT5与肿瘤关系的研究进展

摘    要:信号转导和转录激活因子(STAT)5是STAT家族中的成员,其有STAT5A和STAT5B两个高度同源的亚型。STAT5广泛参与细胞增殖、分化、凋亡、衰老、造血和免疫等多种生物学过程。STAT5失调与白血病、乳腺癌、前列腺癌、黑色素瘤、甲状腺癌等多种恶性肿瘤的发生发展密切相关,且影响肿瘤的放疗、化疗效果以及患者预后。STAT5可作为肿瘤治疗的重要靶点,靶向STAT5可增强前列腺癌、头颈癌等多种肿瘤的放疗敏感性,为肿瘤治疗提供了新的策略。本文就STAT5对肿瘤发生发展的影响、STAT5的靶向治疗策略进行综述。

Abstract:Signal transducer and activator of transcription 5 (STAT5) is a member of signal transducer and activator oftranscription family, which has two highly homologous subtypes: STAT5A and STAT5B. STAT5 is widely involved in variousbiological processes such as cell proliferation, differentiation, apoptosis, senescence, hematopoiesis and immunity. Thedysregulation of STAT5 is closely related to the occurrence and development of malignant tumors such as leukemia, breastcancer, prostate cancer, melanoma and thyroid cancer. STAT5 can also affect tumor radiotherapy and chemotherapy as wellas prognosis of patients. Nowadays, STAT5 has become a key target for cancer treatment. Targeting STAT5 could increasethe radiosensitivity of prostate cancer, head and neck cancer and other tumors, which provided a new strategy for tumortreatment. This article reviews the role of STAT5 in several malignant tumors and strategies of targeting STAT5.
