摘    要:目的:探讨整合素亚基α2(integrin alpha-2,ITGA2)在胰腺癌组织中的表达及作用机制。方法:从GEO基因芯片数据库下载GSE28735的芯片数据分析ITGA2的表达;从TCGA数据库搜索有关ITGA2临床样本表达的数据,分析不同ITGA2表达与胰腺癌临床病理参数的相关性;利用GEPIA在线数据库分析ITGA2对胰腺癌预后的影响;最后,运用基因富集分析探究ITGA2在胰腺癌中的可能作用机制。结果:ITGA2基因在胰腺癌组织中的表达量高于癌旁组织(P<0.000 1),ITGA2基因的表达与胰腺癌患者年龄、T分期、肿瘤的恶性程度G分级具有相关性(P<0.05)。ITGA2高表达胰腺癌患者的总体生存期(P<0.05)和无病生存期(P<0.05)较差。GSEA结果表明:ITGA2基因主要与顶端连接(P=0.021)、E2F转录途径(P=0.036)、mTORC1信号通路(P=0.018)、糖酵解途径(P=0.032)、PI3K/AKT/mTOR信号通路(P=0.028)、G2/M细胞周期检查点(P=0.034)等信号通路有关。结论:ITGA2在胰腺癌组织中表达上调并且与胰腺癌不良的预后相关,其可以作为胰腺癌有效的预后标志物和潜在治疗靶点。

Abstract:Objective:To investigate the expression and mechanism of ITGA2 in pancreatic cancer tissues.Methods:The GSE28735 chip dataset was downloaded from GEO to analyze the expression of ITGA2.The TCGA database was used to search for data on the expression of ITGA2 clinical samples.The correlation between different ITGA2 expression and clinical pathological parameters of pancreatic cancer was analyzed.GEPIA online database was used to analyze the effect of ITGA2 on the prognosis of pancreatic cancer.Finally,gene set enrichment analysis was used to explore the possible mechanism of ITGA2 in pancreatic cancer.Results:The expression of ITGA2 gene in pancreatic cancer tissues was significantly higher than that in normal tissues(P<0.000 1).The expression of ITGA2 gene was correlated with the age,T stage,and G grade of patients with pancreatic cancer(P<0.05).Overall survival(P<0.05) and disease-free survival(P<0.05) were poor in patients with high expression of ITGA2 in pancreatic cancer.GSEA results showed that ITGA2 gene was mainly involved in the apical junction(P=0.021),E2F targets(P=0.036),mTORC1 signaling pathway(P=0.018),and glycolysis pathway(P=0.032),PI3K/AKT/mTOR pathway(P=0.028),G2/M checkpoint(P=0.034).Conclusion:ITGA2 expression is up-regulated in pancreatic cancer tissues and is associated with poor prognosis of pancreatic cancer.It can be used as an effective prognostic biomarker and potential therapeutic target for pancreatic cancer.
