基于Oncomine和GEO数据库荟萃分析HMGA1在卵巢癌中的表达及其与预后的相关性

摘    要:目的:探讨HMGA1在卵巢癌中的表达及其临床意义。方法:对Oncomine和GEO数据库进行检索有关HMGA1的相关信息,并对所获取的信息进行二次分析,并荟萃分析HMGA1在卵巢癌表达中的作用,最后利用GEO数据库中的Kaplan-Meier Plotter进行患者生存周期分析。结果:Oncomine数据库中共收集了449项不同类型的关于HMGA1的研究结果,在肿瘤组织与正常对照组中表达差异具有统计学意义的研究结果共54项,其中HMGA1高表达有48项,低表达有6项。进一步分析发现卵巢癌中HMGA1高表达有8项,低表达0项。这8项研究涉及HMGA1 在卵巢癌组织和正常卵巢组织中的表达,共计355例样本。通过对这8项数据集进行比较分析,发现在卵巢癌组织中HMGA1的表达量较正常卵巢组织显著升高(P<0.05)。通过进一步挖掘GEO 数据库进行Kaplan-Meier分析,我们发现卵巢癌患者HMGA1 的表达水平与卵巢癌和卵巢子宫内膜样腺癌的总生存率(OS)无显著相关性(P>0.05)。结论:我们通过对Oncomine以及GEO基因芯片数据库中肿瘤相关基因信息的深入挖掘,提出HMGA1在卵巢癌组织中高表达,但HMGA1表达量与卵巢癌预后无显著相关性。

Abstract:Objective:To investigate the expression of high mobility group A1 (HMGA1) in ovarian cancer tissue and normal ovarian tissue.And clarify its clinical prognosis significance in ovarian cancer.Methods:Data about HMGA1 were retrieved from the Oncomine database and GEO database,then the prognostic value of HMGA1 was mined and analyzed.Results:For different types of cancer in Oncomine database,there were 449 studies concerning expression of HMGA1,of which 54 studies were of statistically significance (48 up-regulated and 6 down-regulated).A total of 8 of the studies with 355 samples covered the expression of HMGA1 in ovarian cancer tissues and normal tissues.The expression of HMGA1 was significantly higher in the ovarian cancer tissue than in the normal tissues (P<0.05).Furthermore,Kaplan-Meier plots of overall survival indicated that HMGA1 high expression was not tightly associated with overall survival rate (OS) in ovarian cancer (P>0.05).Subgroup analysis revealed that HMGA1 expression was not significantly associated with overall survival rate in ovarian endometrioid adenocarcinoma (P>0.05).Conclusion:HMGA1 is highly expressed in ovarian cancer tissue.However,the expression level of HMGA1 is not significantly correlated with the prognosis of ovarian cancer.
