大鼠脂肪栓塞模型的建立及半数致死量的测定

目的 测定脂肪对大鼠的半数致死量(LD_(50)),建立大鼠脂肪栓塞LD_50模型.方法 静脉注射不同剂量的同种异体大鼠肾周脂肪诱发脂肪栓塞,以死亡率作为评价指标,采用寇氏法计算LD_(50)及其95%可信区间(CI).并取肺、脑、肾组织做冰冻切片,油红O染色和常规苏木精-伊红(H-E)染色.结果 LD_(50)为0.706 mL/kg,其95%CI为0.622～0.801 mL/kg.存活大鼠的冰冻切片的油红O染色中,肺、脑、肾血管内均可见橘红色的脂肪滴,肺部脂肪栓塞相对比较明显,肺小血管、肺组织间隙及肺泡腔内均可见脂肪滴.结论 肾周脂肪的LD_(50)为0.706 mL/kg,由此建立的大鼠脂肪栓塞LD_(50)模型是可靠的.

Establishing fat embolism rat model and determination of half lethal dose

Objective To determine the half lethal dose(LD_(50))of fat in fat-induced embolism and to set up a rat model of fat embolism.Methods Intravenous injection of various doses of perirenal fat of the same generic rats was used to induce fat embolism in rats.The mortality rate was taken as the evaluation index,and Karber's method was used to calculate LD_(50) and the 95% confidence interval.Samples of the lung,brain,and kidney were made into frozen sections for oil red"O"staining or H-E staining.Results The LD_(50) of fat for our animal model was 0.706 mL/kg,with a 95% confidence interval of 0.622-0.80 1 mL/kg.Orange-colored fat droplets were found in the frozen sections of the lung,brain and kidney tissues as stained by oil red"O"staining,with the embolism in the lung being more evident-the fat droplets could be seen in the small pulmonary blood vessels,pulmonary interstitial space,and alveolar cavity.Conclusion The LD_(50) of the perirenal fat for our model is 0.706 mL/kg,and the fat embolism model by intravenous injection of the perirenal fat is reliable.
