摘    要:目的:研究survivin基因转染对人食管癌细胞系KYSE30增殖和侵袭能力的影响。方法:以脂质体将survivineDNA真核表达载体转染人食管癌KYSE30细胞。应用克隆形成实验检测细胞增殖活性的变化,流式细胞仪检测转染前后细胞周期的变化,Transwell小室侵袭实验检测细胞侵袭能力的变化,MTT检测转染前后细胞对各种化疗药物的敏感性。结果:Survivin基因能促进人食管癌KYSE30细胞增殖,流式细胞仪分析结果显示转染后细胞抗凋亡能力增强,侵袭实验显示转染后细胞侵袭能力较转染前有显著增强,MTF实验显示转染后细胞对化疗药物敏感性下降。结论:Survivin基因能够促进人食管癌细胞系KYSE30的体外增殖和侵袭能力,导致对化疗药敏感性下降,提示其对肿瘤的发生发展和细胞耐药起促进作用,可作为食管癌基因治疗的靶标。

Abstract:Objective:To explore the effects of survivin gene on the proliferation and invasion properties of hu-man esophageal squamous cell carcinoma cell line KYSE30. Methods:The survivn cDNA was transfected into human esophageal squamous cell carcinoma cell line KYSE30 by lipofeetamine method. RT - PCR and Western blot assay were performed to detect the level of survivin mRNA and protein. The cloning forming assay was used to evaluate the independent ability of cell anchoring growth. MTF assay was applied to determine the inhibitory rate of cells after drug -pretreated. Flowcytometry was performed to detect apoptosis triggered by VCR. Results:Survivin cDNA could en- hance the growth of esophageal squamous cell carcinoma cell line KYSE30 and their invasiveness, compared with those before transfected. Flowcytometry showed lower apoptosis rate of transfected cells than that of untransfected cells. MTT assay showed that the transfected cells became less sensitive to chemotherapeutic drugs. Conclusion:The results suggested that survivin gene could enhance the abilities of proliferation and invasion in human esophageal squamous cell carcinoma cell line KYSE30 in vitro. These data provided an insight into the probable functions of sur-vivin in generation, progression and drug resistance of human esophageal squamous cell carcinoma, and the survivin may become an effective target of gene therapy.
