阿昔莫司对高脂血症兔主动脉壁脂蛋白脂肪酶表达的影响

摘    要:目的观察阿昔莫司对高脂血症兔主动脉壁脂蛋白脂肪酶表达的影响,探讨阿昔莫司抗动脉粥样硬化的机制。方法健康雄性新西兰大白兔30只,高脂饮食8周后,随机分为高脂组和阿昔莫司组,高脂组(10只),在高脂饮食基础上给予淀粉,阿昔莫司组(10只),在高脂饮食基础上给予阿昔莫司(50mg·kg-1·d-1);正常对照组(10只)给予普通饮食,共14周。采用免疫组织化学染色和半定量分析方法,观察阿昔莫司对主动脉壁脂蛋白脂肪酶表达的影响。结果阿昔莫司组14周后总胆固醇、低密度脂蛋白胆固醇和甘油三酯较高脂组分别下降41.8%、45.9%和46.1%(P均<0.01),高密度脂蛋白胆固醇明显上升(P<0.01);与高脂组相比,阿昔莫司组斑块面积减少1.69倍;阿昔莫司组可显著下调主动脉壁脂蛋白脂肪酶的表达。结论阿昔莫司可能通过下调动脉壁脂蛋白脂肪酶的表达而起到抗动脉粥样硬化作用。

Abstract:Objective To investigate the effects of acipimox on expression of lipoprotein lipase (LPL) on aortic wall in rabbits with hyperlipidemia and the anti-atherogenic mechanisms. Methods Thirty male New Zealand white rabbits fed with 8-week lipid-rich diets were randomly assigned into hyperlipidemia group (n=10) to be fed with starch on the basis of lipid-rich diet for 6 weeks, and acipimox group (n=10) to be fed with aeipimox supplemented with lipid-rich diets (50 mg·kg^-1·d^-1) for 6 weeks. Control group (n=10) was fed with conventional diet for 14 weeks. The effects of aeipimox on LPL expression in aortie wall were determined via immunohistoehemistry assay and semiquantitative analysis. Results Aeipimox produced a decline in total cholesterol, low-density lipoprotein cholesterol and triglyceride by 41.8%, 45.9% and 46.1%, respectively (all P〈0.01). This was associated with an increase in high-density lipoprotein cholesterol (P〈0.01) as compared with hyperlipidemia group. Additionally, aeipimox yielded a 1.69- fold reduction in the surface area of atherosclerotie lesions (P〈0.01) and markedly down-regulated LPL expression in aortic wall. Conclusion The anti-atherogenie mechanisms of aeipimox may be related to down-regulation of LPL in aortic wall.
