黄蘑多糖对H22荷瘤小鼠肿瘤生长及其VEGF表达的影响

摘    要:目的:观察黄蘑多糖对H22(小鼠肝癌细胞系)荷瘤小鼠的抑制肿瘤效果,并探讨该机制是否与提高机体免疫力和抑制肿瘤血管生成有关。方法:采用腋下接种癌细胞悬液建立荷瘤小鼠模型,造模成功24 h后灌胃给药,连续给药12 d后,处死动物,剥离瘤块、胸腺、脾脏并分别称重,计算抑瘤率、胸腺指数、脾指数;眼球取血计数有核细胞数;以冰冻切片免疫组织化学法检测各组瘤组织血管内皮生长因子(VEGF)表达程度。结果:黄蘑多糖各组的肿瘤重量均低于模型组,其中,中、高剂量组的抑瘤效果显著(P〈0.05),且各项免疫指标显著高于环磷酰胺组(CTX组)和模型组(P〈0.01或P〈0.05);黄蘑多糖各组的VEGF表达率显著低于模型组(P〈0.01),黄蘑多糖低、中剂量组VEGF的表达与CTX组比较,差异有统计学意义(P〈0.05),高剂量组与CTX组比较,差异无统计学意义(P〉0.05)。结论:黄蘑多糖对H22移植瘤的生长有一定抑制作用,可降低瘤组织中VEGF的表达,亦可作为生物反应调节剂,通过增强机体免疫功能等作用达到抗肿瘤的功效。
