高原肺水肿动物模型的初步建立

摘    要:
目的:通过模拟高原低压低氧环境,建立高原肺水肿大鼠动物模型,探讨低氧时间对大鼠肺水肿程度影响。方法:雄性Wistar大鼠随机分为3组,分别为对照组、24 h低氧组、48 h低氧组。比较大鼠肺湿干比、肺组织匀浆中一氧化氮合酶(NOS)、内皮素-1(ET-1)及肺组织病理形态学的变化。结果:与地面对照组比较,24 h低氧组与48 h低氧组大鼠肺湿干比及肺组织匀浆内ET-1升高、NOS降低,差异均有统计学意义(均P〈0.05);病理学显示,24 h低氧组与48 h低氧组均有肺泡间隔增宽、间质充血等间质性肺水肿表现,与24 h低氧组相比,48 h低氧组肺湿干比、ET-1升高,肺组织匀浆内NOS降低,差异有统计学意义(P〈0.05)。结论:低压低氧可以有效诱导大鼠产生间质性肺水肿,且48 h低氧大鼠肺水肿程度较24 h低氧更严重,更适合应于现实研究。
