紫杉醇对器官形成期早期小鼠胚胎生长发育的影响

摘    要:紫杉醇剂量设54、18和6mg/kg三个实验组,同时设溶剂对照组、生理盐水对照组和环磷酰胺6mg/kg为阳性对照组。小鼠于孕期d6皮下注射给药,连续给药五d,每d一次。于孕期d11.5处死母鼠,结果表明,紫杉醇各剂量组小鼠早期胚胎的生长发育和形态发育指标与溶剂对照和生理水对照相比均有明显的差别。与给药剂量有关,剂量增加影响更明显。对胚胎组织匀浆生化分析发现54mg/kg组ST和LDH升高,ALP和

EFFECTS OF PACLITAXEL ON THE DEVELOPMENT OF MICE EMBRYOS IN EARLY ORGANOGENESIS PERIOD
Abstract:Paclitaxel 54,18 and 6mg/kg were given subcutaneous injection on gestation mice from 6th day to 11th day.0.9% solution of sodium chloride and polyoxyethylated castor oil,cremphor were used as control and cyclophosphamide was used as a positive control.The mice were sacrificed at 11.5-day of gestation and uterine horns were removed,Implantation sites were counted.embryos were taken off from uterine and examined for malformations and state of development.The results showed that mice embryo growth and development have significant difference compared with control at the 3 dose groups of paclitaxel.Farther experiment showed that in embryo homogenate:AST,LDH increased and ALP,TP significantly decreused at 54mg/kg.ALP,TP decreased and LDH increased at 18mg/kg.ALP decreased and LDH increased at 6mg/kg.
