参附注射液对单侧输尿管梗阻大鼠肾脏肝细胞生长因子及骨形态发生蛋白-7表达的影响

目的探讨单侧输尿管梗阻后大鼠肾间质纤维化发生过程中肝细胞生长因子（HGF）和骨形态发生蛋白-7（BMP-7）的表达以及参附注射液对其的影响。方法采用单侧输尿管结扎（UUO）制造梗阻性肾病模型,将56只大鼠随机分为假手术组、手术组（UUO）和治疗组（UUO＋参附注射液处理）,术后第7、14日观察肾组织病理改变,应用免疫组织化学方法检测肾组织HGF和BMP-7的表达。结果（1）与假手术组相比,手术组肾间质明显纤维化,HGF的表达在术后第7日明显增加,第14日较第7日减弱;与手术组相比,治疗组肾间质纤维化明显减轻,而且HGF的表达在术后第7日明显上调,第14日较第7日上调更明显,差异显著。（2）与假手术组相比,手术组肾间质BMP-7的表达在术后第7、14日逐渐减弱;与手术组相比,治疗组BMP-7的表达在术后第14日明显上调,差异显著。结论参附注射液可以上调肾组织HGF和BMP-7的表达,减轻肾小管-间质纤维化,发挥肾保护作用。

Effect of Shenfu Injection on the Expression of Hepatocyte Growth Factor(HGF)and Bone Morphogenetic Proteins-7(BMP-7)in Rat Kidney after Unilateral Ureteral Obstruction

Objective：To study the expression of hepatocyte growth factor（HGF）and bone morphogenetic proteins-7（BMP-7）during the progression of renal interstitial fibrosis in rat kidney after unilateral ureteral obstruction（UUO）and the effect of Shenfu Injection.Methods：The model was established by UUO,fifty-six rats were randomly assigned into shame operation group,operation group（UUO group）and treatment group（UUO＋Shenfu）.After 7 and l4 days,the renal function and histopathological changes were evaluated.Immunohistochemistry was used to examine the expression of HGF and BMP-7 in renal tissue.Results：①In comparison with the shame opeartion group,the operation group showed renal interstitium fibrosis obviously the expression of HGF increased significantly after 7 days,but then decreased after l4 days.In comparison with the operation group,the degree of interstitium fibrosis was ameliorated obviously in the treatment group,and the expression of HGF increased significantly after treatment for 7 days,even more increased after l4 days.②In comparison with the shame opeartion group,the expression of BMP-7 decreased obviously in the operation group after 7 days,even more decreased after l4 days.In comparison with the operation group,the expression of BMP-7 increased significantly after treatment for l4 day with Shenfu Injection.Conclusion：Shenfu Injection may protect renal by effecting the expression of HGF and BMP-7 in the progression of renal interstitial fibrosis.
