神经节苷脂对脑外伤后脑线粒体呼吸功能和钙泵活性的保护作用

摘    要:目的研究大鼠脑外伤后脑线粒体功能变化及神经节苷脂GM1保护作用。方法从牛脑组织中提取GM1治疗SD大白鼠自由落体脑外伤,测定脑线粒体呼吸功能变化和线粒体、微粒体质膜钙泵活性变化。结果发现脑外伤后8小时和16小时脑线粒体Ⅲ态呼吸耗氧速率、呼吸控制率、磷氧比、氧化磷酸化效率及脑线粒体和微粒体质膜钙泵活性低于正常组,差异有显著性意义(P<0.05或0.01);经GM1治疗后上述指标多数有明显上升,并与正常组相近。结论提示脑外伤后脑线粒体呼吸功能有明显障碍,并与线粒体和内质网钙泵活性下降一致;GM1能显著改善线粒体呼吸功能和提高钙泵活性。

Abstract:Objective In order to investigate brain mitochondrial function changes and GM 1 ganglioside protective effects after traumatic brain injury. Methods GM 1 was isolated from bovine brain tissues to treat experimental free drop brain injury in Sprague-Dawley rats, and the changes of brain mitochondrial respiratory function and Ca 2 -Mg 2 ATPase activity of mitochondria and microsome membrane was determined. Results It was found that R 3 (oxygen uptake rate in states 3 respiration), RCR(respiratory contral ratio),P/O (the molar ratio inorgenic phosphate esterified and oxygen reduced), OPR (oxidative phosphoxylation rate) of brain mitochondria and Ca 2 -Mg 2 ATPase activity of mitochondria and microsome significantly decreased on 8th and 16th hour after traumatic brain injury,while most of these data markly increased after GM 1 given treatment, which were similar to those in normal group. Conclusions These findings suggest that there is severe damage to the respiraory function of brain mitochondria following traumatic brain injury and the damage corresponds to the decrease of Ca 2 -Mg 2 ATPase activity of mitochondria and endoplasmic reticulum. Ganglioside can restore the respiratory function and Ca 2 -Mg 2 ATPase activity.
