柘木镇痛消炎作用实验研究

摘    要:目的观察柘木的镇痛消炎作用。方法取小鼠30只,随机分为空白对照组,罗通定组[60 mg/(kg.d)],柘木高、中、低剂量组[375、250、125 mg/(kg.d],每组各6只。空白对照组予20 mL/(kg.d)蒸馏水灌胃,其余各组予相应药液灌胃,每日1次,连续6 d,采用醋酸致小鼠腹部炎症,观察小鼠扭体次数及潜伏期;取小鼠30只,分组及给药方法、剂量同上,采用热板刺激法造成疼痛小鼠模型,观察小鼠痛阈值。取小鼠30只,随机分为空白对照组,吲哚美辛组[12.5 mg/(kg.d)],柘木高、中、低剂量组(剂量同上),空白对照组予20 mL/(kg.d)蒸馏水灌胃,其余各组予相应药液灌胃,每日1次,连续6 d,观察致炎物对小鼠腹腔通透性的影响;取小鼠30只,分组及给药方法、剂量同上,采用二甲苯致耳肿胀法造成小鼠炎症模型,观察小鼠耳肿胀度。结果与空白对照组比较,罗通定组及柘木高、中、低剂量组小鼠扭体次数均明显减少(P<0.01,P<0.05),罗通定组及柘木高剂量组扭体潜伏期延长(P<0.01,P<0.05),且罗通定组扭体次数少于柘木高、中、低剂量组(P<0.01),柘木高剂量组扭体潜伏期较中剂量组延长(P<0.05)。罗通定组及柘木高、中剂量组小鼠痛阈值与空白对照组比较均明显提高(P<0.01,P<0.05),且罗通定组痛阈值均高于柘木高、中、低剂量组(P<0.05)。吲哚美辛组及柘木高、中、低剂量组OD值与空白对照组比较均明显降低(P<0.01,P<0.05),且吲哚美辛组低于柘木高、中、低剂量组(P<0.05)。吲哚美辛组耳肿胀度与空白对照组比较明显下降(P<0.05),柘木高、中、低剂量组与空白对照组比较有所下降,但差异无统计学意义(P>0.05)。结论柘木具有镇痛消炎作用。

Abstract:【Abstract】 Objective To observe analgesic anti-inflammatory effect of Tsugeki.Methods 30 mouse were randomly divided into five groups,control group,Rotundine group [60 mg/(kg·d)],Tsugeki high-,middle-and low dose group [375,250,125 mg/(kg·d)].Control group received intragastric administration of distilled water [20 mg/(kg·d)].Other groups received intragastric administration of corresponding medicine,once a day for six days.Frequency of stretching and latency period was observed by acetic acid-induced inflammation in mouse abdomen.In another experimental including same grouping,medication and dose,pain threshold of mouse were observed by mouse pain model induced by hot plate stimulation.30 mouse were randomly divided into five groups,control group,indometacin group [12.5 mg/(kg·d)],Tsugeki high-,middle-and low dose group [375,250,125 mg/(kg·d)].Control group received intragastric administration of distilled water [20 mg/(kg·d)].Other groups received intragastric administration of corresponding medicine,once a day for six days.Abdominal permeability in mouse was recorded in each group.In another experimental including same grouping,medication and dose,swelling degree of ear in mouse were observed by xylene-induced ear swelling.Results Frequencies of stretching in Rotundine group,Tsugeki high-,middle-and low dose group were decreased as compared with that in control group(P〈0.01,P〈0.05).Latency period in Rotundine group,Tsugeki high-,middle-and low dose group were prolonged as compared with that in control group(P〈0.01,P〈0.05).Frequencies of stretching in Rotundine group was not so many as that in Tsugeki high-,middle-and low dose group(P〈0.01).Latency period in Tsugeki high dose group were prolonged as compared with that in Tsugeki middle dose group(P〈0.05).Pain threshold of mouse in Rotundine group,Tsugeki high-,and middle dose group were obviously raised in comparison with that in control group(P〈0.01,P〈0.05).Pain threshold of mouse in Rotundine group was higher than that in Tsugeki high-,middle-and low dose group(P〈0.05).OD in indometacin group,Tsugeki high-,middle-and low dose group were significantly decreased as compared with that in control group(P〈0.01,P〈0.05).OD in indometacin group was decreased as compared with that in Tsugeki high-,middle-and low dose group(P〈0.05).Swelling degree of ear in mouse in indometacin group was significantly decreased as compared with that in control group(P〈0.05).Swelling degree of ear in mouse in Tsugeki high-,middle-and low dose group was decreased as compared with that in control group(P〉0.05).Conclusion Tsugeki has analgesic anti-inflammatory effect,375 mg/(kg·d)dose effect was the best.
