外周血单核细胞肺炎衣原体包涵体检测及意义

摘    要:目的探讨外周血单核细胞(PBMC)中肺炎衣原体(Cpn)包涵体的检测方法和临床应用。方法使用低渗溶血法制备PBMC。直接免疫荧光(DIF)检测(Cpn包涵体,以微量免疫荧光(MIF)检测血浆特异性IgG抗体。结果150例冠心病患者Cpn包涵体阳性20例,阳性率13.3%,55例对照组中仅1例阳性,阳性率为1.8%,Cpn IgG两组间阳性率分别为94%及38.2%,冠心病组PBMC中包涵体及血中IgG分别与对照组比较阳性率明显增高(P<0.001)。结论外周血标本来源容易,反映客观。低渗溶血法可集中批量制备PBMC,省时省力。Cpn包涵体阳性反映患者现今Cpn感染状态。

Abstract:Objective The objectives of this study were to search out the method of detecting chlamydia pneumoniae Inclusion bodies in peripheral blood mononuclear cell (PBMC). Methods The PBMC were isolate form 3 ml of the whole blood specimen by low permeate hemolysis method. The presence of Cpn in cell was determined by direct immunofluorescence (DIF) and the serology of chlamydia pneumoniae special IgG antibodies using Microim-munofluorescence(MIF) assay. Results Chlamydia pneumoniae Inclusion bodies was detected in 20(13. 3%) of 150 patients with coronary heart disease( CHD) and in 1(1. 8%) of 55 in control group. Cpn IgG positive rats was 94% in CHD and 38. 2% in control group. The positivity rates in CHD groups were significantly higher than those in control group. Conclusion Chlamydia pneumoniae Inclusion bodies were present in PBMC among the significant proportion of CHD patients. PBMC sample could be collected easily and trustworthily. The positive result in PBMC was showing evidence for the presence of Cpn and was infecting.
