硝苯吡啶防止野百合碱引起大鼠肺动脉高压的研究

摘    要:本文给Wistar系大鼠单次腹腔注射野百合碱50 mg/kg,经过3周引起了肺动脉高压伴有右室肥大。在腹腔注射野百合碱后24h开始灌胃给硝苯吡啶,每天二次,每次5mg/kg、10mg/kg或30mg/kg可显著地减轻野百合碱所致的肺动脉压升高和右室肥大。提示硝苯吡啶长期给药有抑制野百合碱形成肺动脉高压的作用。

Abstract:The influence of nifedipine (Nif) on the development of monocrotaline(MCT) induced pulmonary bypertension was observed in male wistar ratsweighing 100-200 g divided into eight groups.Two groups were given a single dose of saline, two groups were given a single dose of MCT (intraperitonealinjection), three groups received Nif (in PEG-400) 5 mg/kg, 10 mg/kg and30 mg/kg respectively, and one group received PEG-400 (without Nif). Nifand PEG-400 were administered orally bid for 3 weeks, beginning at 24 hoursafter MCT injection. 3 weeks after MCT injection the rats were studied. Itwas found that MCT significantly increased the PAP and RV/LV S; Nifsignificantly reduced the increase in PAP and RV/LV S caused by MCT,PEG-400 did not affect the MCT pulmonary hypertension. These results demo-nstrate that chronic treatment with Nif may reduce the development of MCTpulmonary hypertension.
