逆行示踪法观察甲状腺激素人工神经桥接大鼠坐骨神经缺损的实验研究

摘    要:目的:用逆行示踪方法观察新构建的甲状腺激素人工神经(PDLLA-T3)是否能桥接大鼠坐骨神经缺损。方法:PDLLA-T3连接大鼠坐骨神经1cm缺损后,分别于2周、1个月、2个月三个时相点观察,在神经根远段注射30%辣根过氧化物酶水溶液,3天后收集相应脊髓段进行TMB成色反应,汁数阳性细胞。结果:1个月后在脊髓前脚可见有阳性细胞,平均仅有5—7个,胞体较小。2个月时,实验组阳性细胞增多,每张切片上可以看到大约10个左右的阳性细胞,胞体大,着色深,较正常组少。结论:PDLLA-T3人工神经成功桥接了大鼠坐骨神经缺损,周围神经与神经元胞体问的联系和轴浆运输得到有效恢复。

Abstract:Objective: To apply HRP Retrograde Tracing for the assessment of PDLLA-T3 in rat sciatic nerve injury.Method:After repaired the 1cm rat sciatic nerve defect with PDLLA-T3, 30%HRP were applicated in the distant zone of nerve as assessment of PDLLA-T3 in 2 weeks,1month,and 2months respectively after operation.The corresponding spine were collected,then its slices responsed by TMB,positive cells were calculated,Result:Positive cells were presented in spinal anterior crus after 1 month. There were 5-7 positive cells in each slice,after 2 months,more positive cells appeared,the number increased to 10 in each slice,and more larger in cell body, more denser in color,but less than that in normal. Conclusion: PDLLA-T3 can successfully repaire the defection of rat sciatic nerve,the contaction between peripheral nerve and neuron and the axoplasma flow are effectively recovered.
