耐甲氧西林凝固酶阴性葡萄球菌mecA基因检测分析

摘    要:目的:调查耐甲氧西林凝固酶阴性葡萄球菌(MRCNS)耐药现状,分析其耐药表型与基因型的关系,为临床合理选用抗菌药物提供依据。方法:收集临床分离的86株凝固酶阴性葡萄球菌,采用头孢西丁纸片法检测耐药表型、聚合酶链反应(PCR)检测mecA基因,并将两种结果进行对比分析。结果:86株凝固酶阴性葡萄球菌中头孢西丁纸片法阳性64株,检出率为74.4%;mecA基因阳性58株,阳性率为67.4%;其中有1株头孢西丁纸片法阴性而mecA基因阳性,7株头孢西丁纸片法阳性而mecA基因阴性。结论:本地区耐甲氧西林凝固酶阴性葡萄球菌耐药状况严重;头孢西丁纸片法与PCR检测mecA基因一致率90.7%;PCR检测mecA基因可快速准确判定耐甲氧西林凝固酶阴性葡萄球菌。

Abstract:Objective: To investigate the antimicrobial resistance of methicillin resistance coagulase negative staphylococci (MRCNS)and to analyze the relationship between their phenotype and genotype. Methods: Eighty-six isolated strains of coagulase negative staphylococci(CNS)were collected to detect their drug resistance by cefoxitin disk diffusion method, mecA gene was detected by PCR. Results: Of the 86 strains of CNS, 64 strains showed resistance and 58 strains contained mecA gene. One strain was sensitivity to cefoxitin disk diffusion method but contained a mecA gene and 7 strains were resistant to oxacillin by cefoxitin disk diffusion method but showed negative in mecA gene. Conclusion: The rate of antibiotic resistance of MRCNS is high. By using PCR to detect mecA gene, MRCNS strains can be rapidly determined.
