大肠癌p53基因突变与血清抗体的相关性研究及意义

摘    要:目的:研究大肠癌患者血清p53-Ab与p53基因突变及蛋白表达之间的关系及临床意义。方法:运用ELISA法检测34例大肠癌患者及10例健康人血清p53-Ab,同时运用聚合酶链反应一单链构象多态性分析(PCR-SSCP)和免疫组化(IHC)分别检测p53第5-8外显子突变及蛋白表达状况,结果;大肠癌中血清p53-Ab阳性率为17.6%(6/34),正常对照组血清阴性,P53基因突变率及蛋白表达率分别为52.9%(18/34)和55.9%(19/34),大肠正常粘膜未见P53基因突变及蛋白表达,P53基因突变及蛋白表达与p53-Ab存在及临床病理因素无关(P>0.05),结论:p53基因突变是参与和影响p53蛋白表达的主要因素,P53蛋白表达可诱导p53-Ab产生,但不是唯一决定因素,检测血清p53-Ab有助于大肠癌的诊断及高危人群普查。

Abstract:Objective:To study the relationships between serum p53 antibodies,gene mutation,protein expression and their clinical significances.Methods:p53 antibodies of preoperative sera from 34 patients with colorectal cancer and 10 healthy persons were detected by enzymelinked immunosorbent assay (ELISA).At the same time,by using polymerase chain reaction single strand conformation polymorphism(PCR SSCP)and immunohistochemistry(IHC)S P methods,the mutations in exon 5 8 of p53 gene and protein expression were examined.Rusults:p53 antibodies were positive in 6 of 34 (17.6 %) colorectal cancers,but was not seen in normal serum.The rates of mutations and protein expression of p53 gene were 18 of 34 (52.9 %) and 19 of 34 (55.9 %) in colorectal cancers respectively,no mutation and protein expression were examined in normal mucosa.No relationships were found between p53 antibodies,clinic pathological factors,mutation and expression of p53 gene.Conclusion:Our data indicated that mutation of p53 gene is one of the main factors which participate and affect the protein expression.Protein expression of p53 gene may induce serum p53 antibodies,but not the only factor.Detections of p53 antibodies assist in the early diagnosis and census of high risk population in colorectal cancer.
