清利固精颗粒对系膜增殖性肾炎肾组织PDGFmRNA表达的影响

摘    要:目的:探讨清利固精颗粒对大鼠抗Thy-1系膜增殖性肾炎(MsPGN)肾组织PDGF-A、BmRNA表达的影响。方法:本研究应用大鼠抗Thy-1系膜增殖性肾炎(MsPGN)动物模型,将大鼠随机分为4组:正常对照组、模型组、肾炎康复片组、清利固精组,每组8只大鼠。各实验组于第7天处死大鼠,采用RT-PCR检测肾组织PDGF-A、BmR-NA表达含量。结果:模型组大鼠肾组织PDGP-A、BmRNA表达比正常对照组明显升高(P<0.05~0.01);与模型组相比,清利固精组PDGF-A、BmRNA表达明显下降(P<0.05~0.01);清利固精颗粒降低肾组织PDGF-BmRNA表达疗效优于肾炎康复片(P<0.05)。结论:清利固精颗粒剂抑制细胞增殖及细胞外基质增加,可能与其能降低MsPGN肾组织PDGF-A、BmRNA表达有关。

Abstract:Objective:To investigate the effect of QGG on the expression of PDGF-AmRNA and PDGF-BmRNA in rat kidney tissue with MSPGN.The model was induced by reped injedction of anti-Thy1 antiserum.Methods:32 rats were randomly divided into four groups,the control group,the MSPGN group,the Shenyan Kangfu Tablet(SKT)-treated group and the QGG-treated group.Rats were taken on day 7 to measure on the expression of PDGF-A mRNA and PDGF-BmRNA in rat kidney tissue with RT-PCR.Results:On the expression of PDGF-A mRNA and PDGF-BmRNA in rat kidney tissue in the MSPGN rats as compared with the control rats(P<0.05~0.01).The expression of PDGF-A mRNA and PDGF-BmRNA in rat kidney tissue decrease in the QGG-treated rats compared with the MSPGN rats (P<0.05~0.01).And the effect of decreasing the expression of PDGF- BmRNA in the QGG-treated rats was better than the SKT-treated rats(P<0.05).Conclusion:QGG can inhibit mesangial cell and extra cellullarmatrix proliferation.The mechanism may be it can decreae the expression of PDGF-AmRNA and PDGF-BmRNA in kidney tissue of the MSPGN rats.
