苦参碱联合热疗抗血管生成作用的实验研究

摘    要:目的:探讨苦参碱联合应用热疗对体内外血管生成的抑制作用。方法:采用MTT法观察苦参碱联合热疗对人脐静脉内皮细胞(HUVEC)、人结肠癌LOVO细胞增殖的影响;采用transwell板,观察苦参碱对HUVEC迁移的影响;采用鸡胚绒毛尿囊膜(CAM)模型,观察苦参碱对鸡胚绒毛尿囊膜新生血管的抑制作用。结果:苦参碱在6.25~200μg/ml浓度时,具有抑制HU-VEC增殖的作用(细胞存活率在86.4%~56.1%),与浓度呈负相关((相关系数r为-0.955),此浓度范围内苦参碱对人肠癌细胞株细胞的抑制低于对内皮细胞的抑制,具有明显的差异细胞毒作用;在12.5~25μg/ml浓度时苦参碱对鸡胚尿囊膜血管形成具有显著的抑制作用;在6.25~25μg/ml浓度时对内皮细胞迁移具有显著抑制作用;6.25、12.5、25μg/ml苦参碱联合热疗在体外具有抗血管生成的次加效应。50、100、200μg/ml苦参碱联合热疗在体外具有抗血管生成的协同效应。结论:小剂量苦参碱(6.25~200μg/ml)在体内外具有抑制血管生成作用,联合热疗具有协同或次加效应。

The Experimental Research on the Inhibiting Angiogenesis Effect of Sophorcarpidine With Thermotherapy
Abstract:Objective:To study the effect of sophorcarpidine with thermotherapy on anti-angiogenesis in vitro and in vivo.Methods:We use MTT method to observe the influence of sophorcarpidine on human umbilical vein endothelial cells(HUVEC) or LOVO proliferation.The influence of sophorcarpidine on HUVEC migration was evaluated by transwell.Chick embryo chorioallantoic membrane(CAM) model was used to check whether the neovascularization of CAM could be suppressed in vivo.Result:The survival rate of HUVEC is among86.4%~56.1% within the dose of 6.25~200μg/ml and is negative correlated with the concentration(correlation coefficient is-0.955).The survival rate of LOVO within this doses is more than HUVEC.Sophorcarpidine suppressed angiogenesis of CAM in vivo at doses of 12.5~25μg/ml.and inhibited migration of HUVEC invitro at low doses(6.25~25μg/ml).There is a synergedtic effection in anti-angiogenic when combined use sophorcarpidine(50,100and 200 μg/ml) with thermotherapy in vitro while addition effection whin sophorcarpidine(6.25,12.5and 25 μg/ml) with thermotherapy in vitro.Conclusion: This experiment shows that low dose sophorcarpidine with thermotherapy has anti-angiogenic synergedtic or addition ability both in vivo and in vitro.
