金叶败毒制剂对人巨细胞病毒感染人胚肺细胞的影响

摘    要:目的研究中药金叶败毒制剂对人巨细胞病毒(HCMV)蛋白激酶pul97的抑制作用及对HCMV感染人胚肺细胞病变的影响,探讨其治疗HCMV感染的分子机制。 方法采用半定量逆转录 聚合酶链反应(RT-PCR)方法,检测中药金叶败毒制剂和更昔洛韦(GCV)干预HCMV感染细胞后pul97mRNA表达水平,并观察两种药物干预后对HCMV感染细胞病变的影响。 结果两种药物均可明显抑制HCMV pul97mRNA的表达水平, HCMV感染12 h后, 即出现细胞病变, 而两种药物均可明显延缓感染细胞的细胞病变。 结论金叶败毒制剂通过抑制HCMV蛋白激酶pul97基因的表达而抑制HCMV的表达和复制,从而延缓感染细胞的细胞病变。

Abstract:Objective To study the inhibition of jinyebaidu mixture on human cytomegalovirus(HCMV) protein kinase pul97 in HCMV infection, and to study the cytopathic effect (CPE) after HCMV infection in human embryo fibroblast cells (HEL).The molecular mechanism of HCMV infection was explored. Methods The expression of the HCMV pul97 mRNA in infected cells was measured by semi-quantitative RT-PCR before and after treatment with jinyebaidu mixture or ganciclovir, and the development of cytopathic effect was observed. Results After infection for 12 hours in HEL cells, CPE developed. Both jinyebaidu mixture and ganciclovir showed obvious inhibition in the expression of HCMV pul97 mRNA. And both of them could markedly postpone the development of CPE in the infected cells. Conclusion Jinyebaidu mixture could inhibit duplication of HCMV by inhibiting the expression of HCMV protein kinase pul97. The CPE caused by the HCMV infection can be postponed by jinyebaidu mixture.
