泽泻提取物对高同型半胱氨酸血症兔氧化及抗氧化因子的影响

摘    要:目的:探讨泽泻提取物对高同型半胱氨酸(HHcy)血症家兔血液中部分氧化及抗氧化因子的影响。方法:乙醇回流提取泽泻浸膏。将健康雄性家兔24只随机分为正常对照组、模型组和泽泻组。蛋氨酸皮下注射法建立HHcy血症动物模型,泽泻组灌喂泽泻提取物。8周后检测血浆总同型半胱氨酸(tHcy),血清丙二醛(MDA)、超氧化物歧化酶(SOD)、诱导型一氧化氮(iNO)和一氧化氮合酶(NOS)水平。结果:(1)模型组和泽泻组实验后血浆tHcy明显高于正常对照组,差别有显著性意义(P<0.01);(2)泽泻组血清MDA水平明显低于模型组(P<0.01),泽泻组血清SOD水平明显高于模型组(P<0.05);(3)泽泻组血清NOS和iNO均明显低于模型组(P<0.01)。结论:泽泻可能通过降低iNO活性,抑制过氧化,防止动脉粥样硬化的产生。

Abstract:Objective:To study the mechanism of the extractive from Rhizoma Alismatis influence oxidant/antioxidant factor expression in high homocysteinia rabbits.Method:The effective components in Rhizoma Alismatis was extracted by recirculating alcohol.Twenty-four male rabbits was divided into three groups at random:normal control group(n=4),model group(n=10),therapy group(n=10).Model group and therapy group were given L-methionine subcutaneouse injection very day for eight weeks.At the same time,the therapy group was fed the effective components in Rhizoma Alismatis and the normal control group was given physiologic saline subcutaneouse injection.After 8 weeks,Homocysteine was examined by high performance liquid chromatography(HPLC).Colorimetries were performed to determine superoxided ismutase(SOD),maleic dialdehyde(MDA),superoxide dismutase(SOD),induce-nitric oxide synthase(iNOS)and Nitric oxide synthase(NOS).Results:The level of tHcy in model group and therapy group increased obviously.However,compared with the model group,the level of SOD in the therapy group was higher(P<0.05)and MDA(P<0.01),NOS and iNOS(P<0.01)was lower.Conclusion:These data demonstrated that the effective components in Rhizoma Alismatis can protect the blood vessel endothelium through inhibitting the activity of iNOS,and eliminate the factor of oxygenize.
