Abstract:Purpose To explore the relationship between RASSF7 and epithelial-mesenchymal transition(EMT)of breast cancer in low oxygen conditions.Methods Immunohistochemical staining was used to examine the expression of RASSF7 in 80 pairs of breast cancer tissues and adjacent breast tissues.The relationship of RASSF7 expression with clinicopathological features of breast cancer was analyzed.The expression of RASSF7 in 8 cases of breast cancer tissues and adjacent tissues was detected by Western blot.Western blot was also used to test the expression of RASSF7,HIF-1α,EMT markers of MDA-MB-231 cells in low oxygen conditions.The correlation of RASSF7 and HIF-1α,EMT markers was analyzed by Pearson test.Results The rate of positive expression of RASSF7 in breast cancer tissues(70%,56/80)was significantly higher than that in cancer adjacent tissues(27.5%,22/80)(P<0.05).Western blot results also showed that the expression level of RASSF7 protein in breast cancer tissues was higher than that in cancer adjacent tissues.RASSF7 expression was significantly positively associated to lymph node metastasis and TNM stage in the breast cancer cases(P=0.002 and P=0.027).RASSF7,HIF-1α,vimen-tin and N-cadherin were highly expressed,E-cadherin was lowly expressed in low oxygen conditions.RASSF7 expression was positively correlated with the expression of HIF-1α,vimentin and N-cadherin(r=0.245,P=0.029,r=0.237,P=0.034,r=0.301,P=0.007),and negatively correlated with that of E-cadherin(r=-0.33,P=0.003).Conclusion RASSF7 may induce EMT in low oxygen conditions and promote the invasion and metastasis in breast cancer.
