补脑膏对脑缺血再灌注损伤模型大鼠脑组织VEGF含量的影响

摘    要:目的:观察补脑膏对大鼠脑缺血再灌注损伤后脑组织含水量、组织形态学及脑组织中血管内皮生长因子(VEGF)含量的影响,探讨神经保护作用及机制。方法:雄性SPF级SD大鼠80只,随机分为假手术组8只,模型组、补脑膏大剂量组、补脑膏小剂量组各24只。其中模型组、补脑膏大剂量组、补脑膏小剂量组再分为缺血再灌注24、48和72小时3个亚组,各亚组8只大鼠。除假手术组外,均采用大脑中动脉线栓法制备大鼠脑缺血再灌注损伤模型,再灌注1小时后,灌胃给药补脑膏,2次/d;模型组予生理盐水灌胃。于再灌注72小时末断头取材,制备病理切片进行组织形态学观察,匀浆待测脑组织含水量,ELISA法测定脑组织VEGF含量。结果:1再灌注72小时末,模型组及补脑膏大、小剂量组脑组织含水量较假手术组均升高(P〈0.05);再灌注24、48、72小时各时间点,补脑膏大、小剂量组脑组织含水量较模型组均降低(P〈0.05),而补脑膏大、小剂量组间无明显差异。2再灌注24、48、72小时各时间点,补脑膏大、小剂量组VEGF的含量均较模型组增加(P〈0.05);而补脑膏大剂量组VEGF的含量高于小剂量组(P〈0.05)。3组织形态学改变:假手术组脑组织结构正常。模型组海马区神经细胞排列紊乱、结构模糊,部分细胞变性坏死;补脑膏大、小剂量组海马区神经细胞较模型组集中,变性细胞水肿缓解,细胞排列较有序。结论:补脑膏可通过减轻脑组织水肿及组织形态学改变,增加脑组织VEGF含量而起到脑神经保护作用。

Abstract:Objective:To discuss neuroprotective effects and mechanism of BuNao ointment by observing the effects of BuNao ointment on the contents of VEGF in brain tissue, histomorphology, water content in brain tissue of the rat with cerebral ischemia reperfusion injury. Methods:Eighty SD rats were randomly allocated to eight rats in the sham operation group, 24 rats in the model group, high and low dose groups of BuNao ointment, among them, the model group, high and low dose groups of BuNao ointment were divided into three subgroups:ischemia reperfu-sion for 24 hours, 48 hours and 72 hours, eight rats each subgroup. Except the sham operation group, the rat model suffering from cerebral ischemia reperfusion injury was prepared by middle cerebral artery suture method, in one hour after the reperfusion, the rat model received intragastric administration of BuNao ointment, twice per day;the rats in the model group were administered with physiological saline. In 72 hours after the reperfusion, all the rats were sacrificed to death to prepare pathological slices, histomorphology of the slice was observed, water content of the brain tissue was detected after homogenate, the contents of VEGF in the brain tissue were measured by ELISA method. Results: ①By the end of 72 hours after the reperfusion, the model group, high and low dose groups of BuNao ointment were higher than the sham operation group in water content of the brain tissue (P〈0.05);high and low dose groups of BuNao ointment were lower than the model group in water content of the brain tissue at different times of 24, 48, 72 hours after the reperfusion (P〈0.05), while there was no significant difference between high and low dose groups of BuNao ointment.②High and low dose groups of BuNao ointment were higher than the model group in the contents of VEGF at different times of 24 hours, 48 hours and 72 hours after the reperfusion (P〈0.05);and high dose group of BuNao ointment was higher than low dose group of BuNao ointment in the contents of V
