峨参总黄酮体内外抗氧化作用的研究

摘    要:目的:研究峨参有效部位总黄酮清除羟基自由基和抑制超氧阴离子的作用,以及对D-半乳糖致衰老模型脾脏和肾脏的影响。方法:采用Fenton法产生羟基自由基模型和邻苯三酚自氧化体系,分别测定峨参总黄酮对羟基自由基的清除能力和超氧阴离子的抑制能力;颈部皮下注射D-半乳糖造小鼠衰老模型,研究总黄酮的体内抗氧化作用。结果:峨参总黄酮在27.5-800μg/mL浓度内,清除羟基自由基和抑制超氧阴离子均有较好的量效关系,清除率和抑制率均随浓度的增加而增大,且最大清除率达32.88%,最大抑制率为24.87%;由D-半乳糖造成的小鼠衰老模型,与空白组相比,脾脏和肾脏的模型组MDA含量均升高,SOD活性均降低,有极显著差异(P〈0.05)。结论:峨参总黄酮具有较好的体外抗氧化能力;同时体内抗氧化效果也很显著。

Abstract:Objective:To study eliminating hydroxyl radicals and inhibiting superoxide anion of total flavonoids from EShen Anthriscus Sylvestris (L.).Hoffm], and its influences on the spleen and the kidney of D-galactose inducing aged models. Methods: The properties of eliminating hydroxyl radicals and inhibiting superoxide anion of total flavonoids from EShen were detected respectively by using Fenton method to establish hydroxyl radical models and pyrogallol autoxidation system;antioxidative effect of total flavonoids in vivo was studied by subcutaneous injection of D-galactose inducing aged mice model. Results:Total flavonoids from EShen exhibited better dose-effect relationship in the range between 27.5 μg/mL and 800 μg/mL in eliminating hydroxyl radicals and inhibiting superoxide anion, clearance rate and inhibitory rate were increased as the concentrations were raised, maximum clearance rate reached 32.88%and maximum inhibitory rate 24.87%;D-galactose inducing aged mice model was compared with blank group, the contents of MDA were raised and the activity of SOD decreased in the model group of spleen and kidney, the comparison showed significant difference(P〈0.05). Conclusion:Total flavonoids of EShen possess better antioxidative ability in vitro and remarkable antioxidative ability in vivo at the same time.
