摘 要: | 目的 研究测定人淋巴瘤细胞株Raji摄取~(18)F-氟脱氧葡萄糖(~(18)F-FDG)和~(18)F-氟脱氧胸腺嘧啶核苷(~(18)F-FLT)的方法并比较两者的差异.方法 在不同条件下测定淋巴瘤细胞株Raji对~(18)F-FDG和~(18)F-FLT的细胞结合率:细胞数量1×10~5~1×10~7/瓶;~(18)F-FDG和~(18)F-FLT的放射性活度1.85~29.6 kBq;反应时间20~120min;葡萄糖浓度0~11.1mmol/L.结果 每瓶1×10~7个细胞、加入3.7 kBq ~(18)F-FDG、葡萄糖浓度在0~2.78 mmol/L、作用100min,~(18)F-FDG细胞结合率达到(50.42±1.07)%;每瓶1×10~7个细胞、加入3.7 kBq ~(18)F-FLT、作用100min,~(18)F-FLT细胞结合率达到(59.48±0.77)%;~(18)F-FDG和~(18)F-FLT的细胞结合率之间具有统计学差异(F=1192.805,P<0.001).结论 Raji细胞与~(18)F-FDG的结合率与细胞数量、作用时间及葡萄糖浓度有关,与~(18)F-FDG的放射性活度无关;Raji细胞与~(18)F-FLT的结合率与细胞数量和作用时间有关,与~(18)F-FLT的放射性活度及葡萄糖浓度无关;同等条件下,Raji细胞对~(18)F-FLT的细胞结合率高于~(18)F-FDG.

Abstract: | Objective To investigate the binding characteristics of ~(18)F-fluorodeoxyglucose(~(18)F-FDG)and ~(18)F-fluorodeoxythymidine(~(18)F-FLT)with human lymphoma cell Raji and the differences between them.Methods The binding rates of ~(18)F-FDG and ~(18)F-FLT was measured under different conditions:1×10~5~1×10~7cells.1.85~29.6kBq ~(18)F-FDG or ~(18)F-FLT,0~11.1mmol/L glucose levels and 20~120 min to measure.Results The binding rate of ~(18)F-FDG waa(50.42±1.07)%at the condition of 1×10~7 cells,3.7kBq ~(18)F-FDG,0~2.78mmol/L glucose levels.100min.while ~(18)F-FLT was(59.48±0.77)%at the same conditions.The difiefence of binding rates between~(18)F-FDG and ~(18)F-FLT was significant(F=1192.805,P<0.001).Conclusion The uptake of ~(18)F-FDG and ~(18)F-FLT in human lymphoma Raji cell is high,and at the same condition,the uptake rate of ~(18)F-FLT is higher than ~(18)F-FDG. |
