不同剂量依达拉奉对急性脑梗死患者外周血淋巴细胞DNA损伤的影响

摘    要:目的研究不同剂量依达拉奉对急性脑梗死患者外周血淋巴细胞DNA损伤的影响。方法急性脑梗死患者90例,随机分为3组:观察1组、观察2组和对照组,每组30例。人院后予以常规治疗,观察组在常规治疗基础上每日加用依达拉奉注射液静脉滴注(观察1组:30mg/d,每日1次;观察2组:30mg/d,每日2次)。总疗程7d,观察比较3组患者治疗后外周血淋巴细胞损伤和NIHSS评分。结果观察1组、2组治疗后NIHSS评分均低于治疗前(P〈0.05),且观察1组、2组患者治疗后的NIHSS评分显著低于对照组(P〈0.05)。3组治疗后外周血淋巴细胞DNA损伤均较治疗前减轻(P〈0.05)。在中性单细胞凝胶电泳试验中,观察1组和观察2组患者治疗后外周血淋巴细胞DNA损伤均较对照组减轻(P〈0.05),且观察2组轻于观察1组(P〈0.05);但在碱性单细胞凝胶电泳试验中观察2组与观察1组比较无显著差异(P〉0.05)。结论大剂量依达拉奉保护急性脑梗死外周血淋巴细胞DNA损伤效果更显著,对改善脑梗死预后更具意义。

Effect of edaravone on peripheral blood lymphocyte DNA damage after acute cerebral infarction

Abstract:Objective To investigate the effect of edaravone on peripheral blood lymphocyte DNA damage after cerebral infarct. Methods Ninety acute cerebral infarct patients were randomly divided into observation grouP 1, observation group 2 and control group, 20 patients in each group. All cases were given by general treatment, and the observation groups were additionally given by edaravone (observation group 1:30mg qd, observation group 2:30mg bid). 7 days after treatment, conditions of all groups after the treatment were evaluated by NIHSS, peripheral lymphocytes DNA damage was assessed by single cell gel electrophoresis, and the results were compared. Results NIHSS scores of the treatment group were significant lower than that of the control group after the treatment ( P 〈 0.05). We found that cerebral ischemia resulted in peripheral blood lymphocyte DNA damage of observation group 1 and observation group 2 was significantly attenuated compared with that of the control group in alkaline and neutral electrophoresis ( P 〈 0.05 ). In alkaline electrophoresis, the peripheral blood lymphocyte DNA damage of observation group 2 was no more than that of observation group 1 ( P 〉 0.05). In neutral electrophoresis, the peripheral blood lymphocyte DNA damage of observation group 2 was improved significantly compared with that of observation group 1 ( P 〈 0.05 ). Conclusion Large-dose edaravone can more significantly attenuate the peripheral blood lymphocyte DNA damage in acute cerebral infarction, which might play an more important role on the effect of edaravoneg im- ) proving the prognosis of cerebral infarct
