摘    要:目的探讨siRNA沉默水通道蛋白-4(AQP-4)表达对颅脑损伤(TBI)后创伤性脑水肿的影响。方法90只雄性sD大鼠进行创伤性脑水肿模型制备,随机分为3组,TBI组大鼠不接受任何治疗;AQP-4siRNA组进行siRNA质粒干扰;空载质粒组处理方法同AQP-4siRNA组,但所转质粒不含AQP-4siRNA片段。而后观测大鼠神经功能缺损评分(NIHSS)、脑组织AQP-4mRNA的表达及脑组织含水量。结果治疗后第7、14、21、28天,3组NIHSS均有下降,AQP4siRNA组〈空载质粒组〈TBI组,差异均有统计学意义(均P〈0.05)。治疗后第1、3、5、7天,TBI组、空载质粒组AQP-4mRNA的持续高表达,而AQP-4siRNA组则明显改善(P〈0.05)。造模后3组大鼠脑组织含水量逐渐增加,第3天达到高峰,而后逐渐下降,TBI组与空载质粒组无明显差异(P〉0.05),而AQP-4siRNA组低于TBI组、空载质粒组(P〈0.05)。结论通过siRNA沉默AQP-4表达可明显改善TBI后创伤性脑水肿严重程度,缩短脑功能恢复时间。

Studies on siRNA interferes with aquaporin-4 expression and influences on traumatic brain injury

Abstract:Objective To investigate the aquaporin-4 (AQP-4) effects and mechanism on traumatic brain edema. Methods 90 male SD rats were prepared traumatic brain edema, were randomly divided into three groups, TBI rats received no treatment; AQP-4 siRNA groups with siRNA plasmid interference; empty plasmid group with AQF-4 siRNA treatment group, but that does not contain AQP-4 siRNA transfeeted plasmid fragments. Then observed neurological function in rats (NIHSS), brain tissue AQP-4 mRNA expression and brain water content. Results Mter 7,14,21,28 days treatment, three groups were decreased with NIHSS, AQP-4 siRNA group 〈 empty plasmid group 〈 TBI group, the differences were statistically significant (all P 〈0.05). 1,3,5,7 days after treatment, TBI group, empty plasmid group AQP-4 mRNA continued high expression, whereas AQP-4 siRNA group was significantly improved ( P 〈 0.05 ). After modeling three rats brain water con- tent increased gradually, the first three days reached a peak and then gradually decline, TBI group and empty plasmid group was no significant difference ( P 〉 0.05 ), while AQP-4 siRNA group was lower than TBI group, empty plasmid group ( P 〈 0.05 ). Conclusion By expressing siRNA silencing AQP-4 after TBI can significantly improve the severity of traumatic brain edema, brain function to shorten recovery time.
