摘    要:目的 探讨骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)移植联合电针(electroacupuncture,Ea)治疗对脑出血大鼠脑内巢蛋白(Nestin)表达的影响.方法 取健康SD大鼠110只,随机分为正常组、假手术组、生理盐水组、电针组、BMSCs移植组和BMSCs移植联合电针治疗组.通过向SD大鼠尾壳核注射Ⅰ型胶原酶和肝素制备脑出血模型,并通过BMSCs移植和电针刺激进行治疗,采用免疫组织化学法观察大鼠脑内Nestin的表达.结果 正常组、假手术组、生理盐水组Nestin均有少量表达,三组之间无明显差异.相同时相点电针组、BMSCs组、BMSCs-Ea组Nestin表达依次升高,均高于生理盐水组(P〈0.01);各治疗组出血侧较正常侧高(P〈0.01);在正常侧,同一时相点各治疗组高于对照生理盐水组(P〈0.01).结论 BMSCs移植联合电针治疗脑出血大鼠可上调大鼠脑内Nestin的表达,从而有助于神经组织的修复.

The effects of BMSCs transplantation combined with electroacupuncture on the expression of Nestin in ICH rats

Abstract:Objective To investigate the effects of bone marrow mesenchymal stem cells (BMSCs) transplantation combined with electroacupuncture(Ea) on the expression of nestin in rats subjected intracerebral hemorrhage (ICH). Methods 110 healthy SD rats were randomly divided into normal group, sham operation(SO) group, normal saline (NS) group, Ea group, BMSCs group and BMSCs-Ea group. ICH rat models were established by injecting I collagenase and heparin into caudate putamen, then treated with BMSCs transplantation and Ea, and the expression of Nestin in rat brain was observed by immunohistochemical method. Results The normal group, SO and NS group had little expression of Nestin, and there was no significant difference in the three groups. In comparison with NS group at the same timepoints, the expression of Nestin in Ea group, BMSCs and BMSCs-Ea group increased obviously in turn (P 〈 0. 01 ), and that of the bleeding side of the three groups was higher than normal side (P 〈 0. 01 ). At the normal side, the expression of Nestin in Ea group, BMSCs and BMSCs-Ea group was higher than NS group at the same timepoints (P 〈 0.01 ). Conclusions BMSCs transplantation combined with Ea on ICH rats can increase the expression of Nestin in brain, thus contributing to repair the nerve tissue.
