摘    要:目的 :研究小鼠胚胎着床前后蜕膜组织α 1,3岩藻糖转移酶 (FucT)III~VII型的表达及其在胚胎着床中的作用。方法 :取未孕和孕 1~ 7d小鼠子宫内膜 ,每组 6例。用RT PCR法测定III~VII型mRNA水平的表达 ,并经测序鉴定。结果 :正常小鼠内膜有一定的FucT IV的表达 ,其表达在妊娠第 1天和胚胎着床前后即妊娠第 4天出现峰值。结论 :5种糖基转移酶中只有FucT IV参与了胚胎着床的过程。它的作用可能是参与了胚泡表面特异性抗原Lex等抗原的糖基化 ,从而促进胚胎的植入。

Abstract:Objective:To study the expression of α1,3 fucosyltransferase(FucT),the key enzymes responsible to synthesize the sLe x antigen,in order to reveal their relationships to the embryo implantation.Methods:The endometrium of mouse during the day 1 7 th pregnancy was collected for measurement of 5 types of α1,3 FucT.Nonpregnancy mouse acted as control group.The expression of α1,3 FucT mRNA was determined by RT PCR method.DNA sequencing was used to identify the PCR product.Results:The qualitative and quantitative analyses of the mRNA expression of α1,3 FucT subtypes were carried out in the case of embryo implantation.The length of the amplified PCR products for α1,3 FucT IV was 318bp same as the fragments theoretically determination.DNA sequencing showed that the amplified fragment with 318bp has 98% homogeneity to the cDNA of IV type.The peak expression of FucT IV mRNA was on the day 1 and 4 of pregnancy,which were higher than those in normal endometrium,with significant difference (P<0.05).Conclusion:In the five subtypes,only FucT IV is involved in the process of embryo implantation.FucT IV may affect the adhesion between the embryo and endometrium.
