芪加真武汤对免疫抑制小鼠淋巴细胞转化及血清白介素2的影响

摘    要:目的研究芪加真武汤颗粒对免疫抑制小鼠的作用,旨在探讨其作用机制,为临床使用提供实验依据。方法将实验动物(昆明种小鼠)随机分为4组,每组10只。实验组:环磷酰胺(CTX)+芪加真武汤。阳性对照组:CTX+胸腺肽。病理模型组:CTX+生理盐水(NS)。正常对照组:NS+NS。实验组小鼠用芪加真武汤灌胃4周。第3周末前三组用环磷酰胺150 mg/kg一次性腹腔注射,造成免疫抑制模型。第4周断头取血,应用双抗夹心ELISA法检测四组小鼠血清白细胞介素-2(IL-2)水平。用3H-TdR渗入法检测小鼠脾脏淋巴细胞转化率。结果与正常对照组比较,病理模型组IL-2水平降低(P<0.01),淋巴细胞转化率及血清溶血素均降低(P<0.05)。与病理模型组比较,实验组显著提高IL-2(P<0.01)、淋巴细胞转化率(P<0.01)及血清溶血素水平(P<0.05)。与阳性对照组比较,实验组IL-2、淋巴细胞转化率无统计学意义(P>0.05)。结论 芪加真武汤颗粒能显著提高免疫抑制小鼠淋巴细胞转化率及血清IL-2水平。

Abstract:Objective To investigate the effect of Qijia Zhenwu Tang granules (QJZWT) on immunodepressive mice. Aimed at researching its working mechanism, and providing the experimental basis for clinical application. Methods Experimental animals (Kunming mouse) were divided randomly into four groups, that is, Experimental group: Cyclophos-phamide(CTX)+QJZWT, Positive control group: CTX+Thymosin, Pathologic model group: CTX+Saline (NS) and Normal control group: NS+NS. Each group had ten mice. Each group had been treated with QJZWT, Thymosin or NS for four weeks. The animals of the former three groups were injected CTX (150 nig/kg) into the abdominal cavity to induce immunodepressive model at the end of third week. At the end of fourth week, all mice were killed, and mice serum inter-leukin-2(IL-2)was tested by BS-ELISA, the transformation rate of lymphocyte was measured by 3H TdR. Results Compared with normal group, serum IL-2 (P<0.01), serum hemolysin and the transformation rate of lymphocyte(P<0.05) depressed significantly in pathologic model group. In comparison with pathologic model group, IL-2(P<0.01), the transformation rate of lymphocyte(P<0.01) and serum hemolysin (P<0.05) increased significantly in experimental group. There was no statistical difference between positive control group and experimental group on IL-2 and the transformation rate of lymphocyte (P>0.05). Conclusion This experiment proved that QJZWT could promote the transformation rate of lymphocyte and the content of serum IL-2 of immunodepressive mice.
