羊膜移植治疗急性角膜碱烧伤的实验研究--基质金属蛋白酶及其抑制剂表达的测定

摘    要:目的 探讨新鲜羊膜移植术在早期角膜碱烧伤治疗中的作用机制。方法36只新西兰大白兔制作角膜碱烧伤模型,分为新鲜羊膜移植组、保存羊膜移植组和对照组。术后每日观察新生血管生长情况,并于术后7天、14天应用计算机图像分析系统对角膜表达基质金属蛋白酶(MMPs)和基质金属蛋白酶抑制剂-1(TIMP-1)进行半定量测定。结果术后新鲜羊膜移植组角膜新生血管发展缓慢,MMFs角膜表达降低而TIMP-1表达升高。结论羊膜移植可通过提高角膜表达基质金属蛋白酶抑制剂-1的水平来抑制基质金属蛋白酶的活性,从而在早期角膜碱烧伤的治疗中起抑制角膜溃疡和新生血管形成的作用,且这一作用新鲜羊膜要明显优于保存羊膜。

Abstract:Objective To investigate the mechanism of fresh amniotic membrane transplantation on inhibiting corneal ulcer and restraining neovascularization in early corneal alkali burn.Methods Corneal alkali-burn model of 36 New Zealand white rabbits was made.The rabbits were divided into fresh amniotic membrane transplantation group (fAMT),preserved amniotic membrane transplantation group (pAMT) and control group (C).On the 7th day and 14th day after operation,the expression of MMP-1,MMP-9,TIMP-1 in cornea were measured by using a computer image analysis system.Results The levels of MMP-1 and MMP-9 in fAMT were decreased significantly when compared with pAMT,and both of them were lowr than those in C.The expression of TIMP-1 in fAMT were increased when compared with pAMT,and both of them were higher than those in C.At the same time,the levels of MMP-1,MMP-9 both had significant negative correlation with the levels of TIMP-1.Conclusion Fresh amniotic membrane transplantation has stronger effect on inhibiting expression of MMP S than that of preserved amniotic membrane transplantation in early corneal alkali burn.
