阿托伐他汀对大鼠缺血再灌注脑组织中CD40L和MMP-3的抑制作用

摘    要:目的:探讨阿托伐他汀对脑缺血再灌注损伤的脑保护作用机制。方法:采用Zealonga线栓法制作大鼠大脑中动脉缺血再灌注模型血管内皮细胞间黏附分子。将Wistar大鼠随机分组:假手术组(Sham组)、缺血再灌注组(IR组)、阿托伐他汀治疗组(AT组)。采用免疫组织化学法检测大鼠脑组织CD40L、金属基质蛋白酶-3(MMP-3)的表达。结果:AT组脑组织CD40L、MMP-3表达及脑含水量和神经功能评分较IR组低(P〈0.01),而较Sham组明显增高(P〈0.01)。结论:阿托伐他汀可以通过抑制大鼠脑缺血再灌注后脑组织CD40L、MMP-3表达,发挥脑保护作用。

Abstract:Objective:To study the neuroprotective mechanism of atorvastatin against the injury induced by cerebral ischemia-reperfusion.Methods:The middle cerebral artery was occluded with thread according to modified Zealonga′s method.Wistar rats were randomly divided into:sham-operation group(Sham group),ischemia-reperfusion group(IR group),atorvastatin treatment group(AT group).The expressions of CD40L,MMP-3 in brain tissues were measured by immunohistochemical methods.Results:The expressions of CD40L,MMP-3,neurologicol scores,subtract brainedema in AT group were lower than those in IR group(P0.01),significantly higher than those in sham group(P0.01).Conclusion:Atorvastatin can protect neural function by downregulating CD40L、MMP-3 expressions during focal cerebral ischemia-reperfusion.
