EB病毒BNLF-1基因在淋巴瘤和急性白血病的表达及临床意义

摘    要:目的:研究EB病毒BNLF-1基因在淋巴瘤和急性白血病的表达及含量,探讨对该基因表达与淋巴瘤、急性白血病的发病机制与预后的关系。方法:采用Amplisensor荧光定量PCR技术,检测淋巴瘤和急性白血病患者血液或骨髓EB病毒BNLF-1基因片段,同时测定血清中EB病毒VCA-IgA抗体滴度。结果:(1)淋巴瘤BNLF-1基因表达高于急性淋巴细胞白血病(急淋)、急性非淋巴细胞白血病(急非淋)和对照组;急淋高于急非淋和对照组;淋巴瘤VCA-IgA阳性率高于急非淋和对照组;急淋高于对照组;急非淋与对照组无显著差异。(2)淋巴瘤BNLF-1基因含量高于急淋、急非淋;急淋高于急非淋。(3)T、B细胞淋巴瘤BNLF-1基因表达无显著差异。(4)淋巴瘤BNLF-1基因含量越高,生存期越短。结论;BNLF-1基因可能是EBV的致癌基因之一;其阳性率及含量的高低可作为淋巴瘤及急淋和急非淋的鉴别诊断依据之一;BNLF-1基因高表达与淋巴瘤的预后呈负相关关系。

Abstract:Objective To study the expression and quantity of Epstein|Barr virus(EBV) BamHI|N leftward reading fragment(BNLF|1) gene fragment and its clinical significance in lymphoma, acute lymphocytic leukemia(ALL), and acute non|lymphocytic leukemia(ANLL). Methods The BNLF|1 gene fragment was amplified and quantified by using quantitative polymerase chain reaction(PCR), and VCA|IgA was detected by enzyme immunoassay. Results (1)The positive expressive rate of BNLF|1 was higher in lymphoma than that in ALL, ANLL, and the control. It was higher in ALL than that in ANLL and in the control. The positive rate of VCA|IgA was higher in lymphoma than that in ALL and the control. It was higher in ALL than that in the control. There were no significantly difference between {ANLL} and the control. (2)The quantity of BNLF|1 was higher in lymphoma than that in ALL and {ANLL}. It was higher in ALL than that in ANLL. (3)There were no significantly difference of expressive rate and quantity of BNLF|1 gene between T cell and B cell lymphoma. (4)It showed negative correlation between the quantity of BNLF|1 gene and life span in lymphoma. Conclusion BNLF|1 may be one of carcinogens of lymphoma and acute lymphocytic leukemia. BNLF|1 may serve as one of the criteria for differential diagnosis of lymphoma, ALL, and ANLL. The higher quantity of BNLF|1 gene, the shorter life span is.
