降脂通脉方对模型家兔主动脉平滑肌细胞低密度脂蛋白受体相关蛋白mRNA表达的影响

目的探讨降脂通脉方对食饵性动脉粥样硬化家兔主动脉平滑肌细胞低密度脂蛋白受体相关蛋白(LRP)mRNA表达的影响。方法将24只家兔随机分为空白对照组、动脉粥样硬化模型组、降脂通脉方组、血脂康药物对照组。采用胆固醇高脂饮食喂养复制家兔动脉粥样硬化模型,60d后开始灌胃给药,30d后处死取主动脉平滑肌。采用RT-PCR检测LRPmRNA表达。结果模型组与空白组比较,主动脉平滑肌细胞LRPmRNA表达水平均明显增高(P<0.01);降脂通脉方能显著降低动脉粥样硬化家兔主动脉平滑肌细胞LRPmRNA表达(P<0.01)。结论降脂通脉方能使动脉粥样硬化家兔主动脉平滑肌LRPmRNA表达下调,提示降脂通脉方可能通过调节家兔主动脉平滑肌细胞LRP来防治动脉粥样硬化。

The Effect of Jiangzhitongmai Receptu on the Low Density Lipoprotein Receptor-related Protein mRNA Expression on the Aortic Smooth Muscle Cells in the Rabbits with Food-induced Atherosclerosis

Objective To explore the pharmacological mechanism of Jiangzhitongmai Receptu against atherosclerosis by the low density lipoprotein receptor-related protein(LRP)mRNA expression on the aortic smooth muscle cells.Methods AS animal models were induced by feeding lipid-rich food within 60 days.Jiangzhitongmai Receptu was taken from the 61st day to the 90th day.At the end of the experiment,the LRP mRNA expression in the aortic smooth muscle cells(SMC)was detected by using RT-PCR technique.Results The LRP mRNA expression levels of SMC inthe model group were significantly higher than that in control group(P <0.01).Jiangzhitongmai Receptu could decrease the expression level of LRP mRNA,with very significant difference when compared with that of the model group(P <0.01).Conclusion Jiangzhitongmai Receptu can down-regulated the LRP mRNA expression in the induced AS animal models,which implied that Jiangzhitongmai Receptu might prevent the occurrence of AS by regulating the SMC LRP mRNA expression.
