异种抗原治疗肿瘤对荷瘤小鼠调节性T细胞的影响

摘    要:目的研究异种抗原免疫续贯瘤内注射的方法对荷瘤小鼠肿瘤局部调节性T细胞数量的影响,及其对肿瘤的治疗作用。方法利用异种抗原免疫续贯瘤内注射的方法处理试验小鼠,测量瘤块大小,计算抑瘤率,使用流式细胞术检测肿瘤主要组织相备性合体Ⅰ类(MHC-Ⅰ)类型表达及调节性T细胞的数量,使用ELISA方法检测瘤内白细胞介素2(IL-2)浓度。结果实验组小鼠的MHC-Ⅰantigen、CD3 CD8 T细胞以及CD3 CD4 CD25 T细胞的表达率分别为42.45%、22.58%和2.91%,IL-2的浓度为100.6pg/ml,与免疫对照组和空白对照组之间存在显著性差异(P<0.01)。结论异种抗原免疫续贯瘤内注射的治疗方法可以上调肿瘤MHC-Ⅰ类抗原,减少肿瘤局部调节性T细胞的数量,有良好的抗肿瘤作用。

Abstract:Objective The therapeutic effect of sequential intratumoral injection with xenogenic antigens after immunization on the mice with tumor and its relationship with the number of regulatory T cells in tumor tissue is studied in this paper.Methods The treatment method of xenogenic antigen immunization sequential with intratumoral injection has been performed.The tumor sizes of mice are obtained,as well as the tumor-inhibition ratios.The CD3 CD4 CD25 Treg cells and the tumor MHC-I antigen presentation are counted by using Flow Cy-tometric Analysis.The concentration of IL-2 in tumor tissue is also obtained by means of ELISA.All the experimental data from different groups are compared.Results In the experimental group,the proportions of the tumor MHC-I antigen,the ratio of CD3 CD8 T lymphocytes and the CD3 CD4 CD25 Treg cells are 42.45%,22.58% and 2.91%,respectively.The concentration of IL-2 is 100.6 pg/ml,which is signifi-cantly different from the control groups(P < 0.01).Conclusion The combined treatment of intratumoral injection with xenogenic antigen and the immunotherapy is an effective anti-tumor method.It can significantly increase the amount of effector cells in the tumor micro-environ-ment,and decrease the expression of Treg lymphocytes.
