庆大霉素对豚鼠耳蜗柯替器毛细胞凋亡作用的影响

目的 研究庆大霉素对于豚鼠耳蜗柯替器毛细胞的凋亡作用。方法 将豚鼠分为生理氯化钠溶液对照组和庆大霉素组，连续给药14d，利用TUNEL法原位检测动物耳蜗柯替器毛细胞的凋亡情况。结果 生理氯化钠溶液对照组豚鼠耳蜗未见凋亡柯替器毛细胞，庆大霉素组中耳蜗基底回柯替器内外毛细胞均消失，第三回中第1排外毛细胞消失，第2、3排外毛细胞均呈阳性表达，第二回中第1、第2排外毛细胞呈阳性表达。结论 庆大霉素作用于豚鼠耳蜗时可引起柯替器毛细胞凋亡，细胞凋亡是豚鼠耳蜗柯替器毛细胞损伤的一条途径；庆大霉素耳毒性的产生可能与其引起耳蜗柯替器毛细胞凋亡有关。

Apoptosis of Guinea Pig Cochlear Corti's Hair Cells Following Gentamicin Treatment

Aim To study the effect of Gentamicin on apoptosis of guinea pig cochlear Corti's hair cells. Methods Twenty-four guinea pigs were randomized to the normal saline group and Gentamicin group. After 14 days' successime administration, apoptosis of their cochlear Corti's hair cells was detected by TUNEL method in situ. Results In the normal saline group apoptosis of Corti's hair cells as absent, but in the Gentamicin group the disappearance of OHCs and IHCs was observed in the Corti of cochlear basal turn and OHCs in the first row of the third turn disappeared, Labeled nuclei could seen in OHCs in both the second row of the third turn and the third row of the third turn, as well in OHCs in the first and second row of the second turn. Conclusion Gentamicin can induce the apoptosis of cochlear Corti's HCs. Ototoxicity induced by Gentamicin may be related to apoptosis of cochlear Corti's HCs.
