丙戊酸镁对利培酮疗效及血药浓度的影响

目的观察小剂量丙戊酸镁对利培酮疗效，不良反应及血药浓度的影响。方法将60例精神分裂症患者随机分为对照组（利培酮组）30例和研究组（利培酮＋丙戊酸镁）30例。对照组单纯使用利培酮治疗，限定治疗剂量为4mg；研究组同时联合丙戊酸镁片（500-750mg／d）。分别于治疗前与治疗2周末、治疗4周末采用阳性与阴性症状量表（PANSS）评定疗效。采用不良反应量表（TEss）评定治疗中不良反应。于治疗2周末、4周末测定利培酮血药浓度。结果两组利培酮血药浓度测定结果在治疗2周末、4周末比较差异无显著性（P〉0．05）。PANSS评分结果显示，在治疗2周末，研究组与对照组相比，差异有显著性（P〈O．05），而4周末的差异无显著性（P〉0．05）。两组TESS评分结果比较差异无显著性（P〉0．05）。结论联用小剂量丙戊酸镁不会增加利培酮血药浓度。疗效与血药浓度无明显相关，其不良反应未受影响。

Effect of magnesium valproate on risperidone efficacy and serum drug concentration

Objective Clinical observation of small dose of magnesium valproate on risperidone efficacy, adverse reactions and the effect of drug concentration in blood plasma. Methods 60 Patients with schizophrenia were ran domly divided into control group （30 cases of risperidone group） and study group （risperidone plus magnesium val proate） in 30 cases. The control group was treated with risperidone, defining therapeutic doses for 4mg; study group at the same time combined with magnesium valproate tablets （500-750mg/d）. In treatment 2 weekend, 4 weeks after the treatment using positive and negative symptom scale （PANSS） assessment of efficacy. The adverse reaction scale （TESS） assessment of adverse reactions during treatment. In the treatment of 2 weekend, 4 weekend determi nation of the plasma concentration of risperidone. Results 2 Groups of the plasma concentration of risperidone in treatment of determination results of 2 weekend, the 4 weekend of no significant difference （ P 〈 0.05）. The score of PANSS results show： in the treatment of 2 weekend, the study group compared with the control group, the difference was statistically significant （ P 〈0.05）, while the 4 weekend of the differences were not statistically sig nificant （ P 〈0.05）. 2 groups of TESS score was no significant differences. Conclusion Combined with small dose of valproic acid magnesium does not increase the plasma concentration of risperidone, curative effect and serum con centration was not significantly related to, the adverse reaction of unaffected.
