康复训练对脑缺血大鼠神经功能恢复和脑组织中白介素10含量变化的影响

摘    要:目的 探讨康复训练对脑缺血大鼠神经功能恢复和脑组织中白介素-10(interleukin-10,IL—10)含量变化的影响。方法 采用改良Zea—Longa线栓法将36只SD大鼠制作成大脑中动脉栓塞(MCAO)动物模型。并于48h后将其随机分成康复组和对照组。康复组大鼠每天给予平衡、抓握、旋转、行走等运动功能训练。对照组则置于普通笼内饲养。期间可自由活动、进食。2组大鼠分别于造模后3,7及14d时进行神经功能评估,并断头取脑组织制成标本。采用酶联免疫吸附分析法(ELISA)测定脑组织中IL-10的水平。结果 康复组大鼠MCAO后7d时和14d时的神经功能评分均明显优于对照组;康复组大鼠MCAO后7d时和14d时脑组织中IL-10浓度亦均较对照组明显增高。结论 运动康复训练可刺激脑缺血大鼠脑组织中IL-10生成,这可能是运动康复训练促进脑缺血大鼠神经功能恢复的重要机制之一。

Abstract:Objective To study the effects of rehabilitation on the recovery of neurological functioning and interleukin-10 levels in the brain post focal cerebral ischemia in rats. Methods Permanent focal cerebral ischemia was induced by right middle cerebral artery occlusion ( MCAO) in 36 male adult Sprague-Dawley rats using a modification of Longa's method. The rats were rats were randomly divided into 2 groups 48 hours post-MCAO: a rehabilitation group, in which the rats received balancing, grasping, rotating and walking training every day; and a control group, in which the rats could move freely in standard cages. Their neurological functions were measured at days 3, 7 and 14 post-MCAO. They were then sacrificed, and the concentration of interleukin-10 in their brains was determined by the ELISA method. Results The rats in the rehabilitation group showed significantly better neurological functioning 7 d and 14 d post-MCAO (P <0.05) , and significantly higher interleukin-10 concentration (P<0.05) in their brains, as compared with the control group. Conclusion Rehabilitation may promote recovery of neural function and interleukin-10 synthesis in the brains of rats with permanent MCAO.
