葛根总黄酮和葛根素的纯化工艺研究

摘    要:目的研究葛根总黄酮和葛根素的纯化工艺。方法以葛根总黄酮和葛根素为指标,对萃取法、盐析法、活性炭吸附法以及大孔树脂吸附法四种纯化方法进行综合评价,并对大孔树脂吸附法进行了工艺优化。结果四种纯化方法中,大孔树脂吸附法效果最佳。非极性大孔树脂更加有利于葛根总黄酮及葛根素的纯化,且以HPD 300最佳。结论HPD 300树脂吸附纯化时,以上样液浓度1g.mL-1(相当于原生药),20%乙醇洗脱效果最佳,其中葛根总黄酮可达90%以上,葛根素纯度可达50%以上,能满足大工业生产的要求。

Abstract:OBJECTIVE To study the proper process for purifying Pueraria isoflavones and puerarin.METHODS With the content of the total isoflavones and puerarin as detective markers,the process was studied by comparing 4 purification methods,and optimizing purification with macroporous resin.RESULTS Purification with macroporous resin is better than other methods,and the HPD300 is the best choice for purifying Pueraria isoflavones and puerarin.CONCLUSION When using HPD300 with the flavones content in the liquid is 1.0g per 1 ml(equivalemt to raw material) and 20% ethanol as eluant,the total flavones' purity could reach 90% and the puerarin's purity could reach 50%.Therefore,the process can meet the industrial requirements.
