前列腺增生组织双氢睾酮结合容量的研究

摘    要:目的 探讨组织双氢睾酮(DHT)结合能力在良性前列腺增生(BPH)病因中的作用。方法32例BPH患者的前列腺组织取自耻骨上经膀胱前列腺摘除术,制备组织胞浆、胞核提取液,以乙醚萃取的方法去除提取液中所有的内源性类固醇激素,加入3H-DHT测定结合的3H -DHT的含量。结果 BPH组织中DHT结合蛋白的总DHT结合容量均数约为0.024nmol/g湿组织,胞浆、胞核DHT结合容量分别为(0.01280.0020)nmol/g湿组织和(0.01120.0059)nmol/g湿组织,两者比较差异无显著性(P>0.05)。结论 BPH组织中的DHT结合蛋白具有较高的DHT结合容量,维持局部DHT 的高浓度,有利于DHT在前列腺组织增生过程中的作用。

Abstract:Objectives To quantify DHT-binding ability and to study the effect of the DHT-binding ability onthe etiology of the BPH. Methods Cytosolic and nuclear fractions was obtained from 32 BPH tissues by superpubicprostatectomy, and all the endogenous hormone were removed from the cytosolic and nuclear fractions by etherstripping. To add 3H-DHT, and the content of the bound 3H-DHT was assayed. Results The average DHT-bindingcapacity of the DHT-binding protein in BPH is 0.024nmol/g wet tissue. The DHT-binding capacity of cytosolic andnuclear fractions are (0.01280.0020)nmol/g wet tissue and (0.01120.0059)nmol/g wet tissue respectively, the differ-ence of the two capacities is not significant (P>0.05). Conclusions The DHT-binding capacity of the DHT-bindingprotein in BPH is high and maintains the high DHT level and facilitates the effect of DHT on BPH.
