Abstract:Objective To study genetic structure and phylogeny of the Tujia and Han populations in Hubei province, China, and to discuss their origins and relationships with other populations in China. Methods Based on the fingerprints of 15 short tandem repeats (STR) loci, the genetic diversity and clustering analyses were conducted with 333 cases of Hubei Tus and 89 cases of Hubei Hans as population samples, which have no direct relationship with each other and belong to the same population in ascendant three generations. Multidimensional scaling (MDS) analysis was also conducted to reveal the genetic relationship between the Tujia and the Han populations. Results The STR fingerprinting technique had high resolution on both Hubei Tujia and Hubei Han populations, and could be broadly used in individual recognition, forensic identification and population genetics researches on these populations. The core group of the Tujia consisted of subgroups Ⅰ (8.8%) and Ⅱ (33.9%). MDS analysis revealed no significant divergence between the core groups of the Tujia and the Han populations in Hubei. The similarity of STR genotype frequency between Hubei Tujia (North subdivision of the Tujia) and Chongqing Tujia (South subdivision of the Tujia) populations was 91.2%, whereas it was 95.3% between Hubei Tujia and Hubei Han populations. Although Chongqing Tujia was significantly divergent from the Han, Miao and Yao populations, this group was genetically close to the Lahu and Bai populations in Yunnan province with a similarity of 92.1%. Conclusions The tight genetic relationship between the Tujia and Han populations in Hubei indicated that broad genetic exchanges have occurred among them in history. The genetic divergence between Hubei Tujia and Chongqing Tujia populations reflected that the Tujia population in China has significantly split during the process of evolution.
