硒对氟所致细胞凋亡及p53基因的影响

摘    要:目的 研究硒对慢性氟中毒大鼠肝肾细胞凋亡及相关基因的影响。方法 对饮高氟水(100mg/L)及高氟加不同浓度硒饲料(2.3mg/kg)大鼠饲喂4个月后,取其肝肾组织进行透射电镜形态学观察、DNA凝胶电泳及流式细胞术观察各组细胞凋亡情况,同时检测肝肾细胞凋亡的相关基因p53。结果 与对照组相比,高氟及氟加硒组.肝肾细胞凋亡率及p53显著增加,氟加硒组肝肾细胞凋亡率明显低于单纯加氟组。结论 高氟可诱导细胞凋亡,硒可拮抗氟引起的细胞凋亡,且p53参与了细胞凋亡的调控。

Abstract:Objective To study the effect of Se on apoptosis of rats caused by high flouride in drinking water. Methods Using techniques of flow cytometry, DNA agarose gel electrophoresis and transmission electronic microscope(TEM), we investigated the effect of high fluoride and high fluoride adding Se on apoptosis in hepatocyte and kidney. P53 was detcted in hepatocytes and kidney by using flow cytometry. Result Hepatocyte and kidney apoptosis were obviously increased in high fluoride group and the groups of adding Se, as compared with the control group. P53 gene was evidently increased too. But apoptosis of Se supplemented groups were lower than that of the group of only adding fluoride. Conclusion High fluoride exposion can induce hepatocyte and kidney apoptosis in rats. Se could antagonize high fluorosis, and decrease the occurrence of apoptosis.
