糖尿病大鼠视网膜色素上皮细胞超微结构

摘    要:目的 :探讨糖尿病大鼠视网膜色素上皮细胞超微结构病变 ,为糖尿病视功能障碍提供有关的行态学依据。方法 :选择清洁级SD大鼠随机分成正常对照组、糖尿病 1月、3月和 6月。腹腔内注射链脲佐菌素 (STZ)诱导大鼠糖尿病 ,每组 1 2只 ,取视网膜制备超薄切片 ,透射电镜观察。处死前每月测体重、血糖 1次。结果 :糖网病大鼠视网膜色素上皮细胞的微绒毛和基底部的质膜内褶随血糖的增高病情加重而逐渐稀疏、低矮直至脱落、低平。胞质内细胞器以线粒体的脱嵴、水肿和内质网的扩张为主要损害 ,而相邻色素上皮细胞之间的连接复合体始终存在。结论 :糖网病大鼠视网膜色素上皮细胞的损害主要表现于微绒毛、质膜内褶及细胞器的损害 ,并无脉络膜与视网膜之间屏障功能的损害。本研究为糖网病的临床研究提供形态学依据

Abstract:Objective:To probe into the ultrastructural changes of retinal pigment epithelium in diabetic rats providing the concerning morphological evidence for visual functional disorder in diabetic retinopathy. Methods:SD rats were induced by injected streptozotocin (60 mg/kg) into abdominal cavity,and were randomly divided into normal group (CON),diabetic groups 1 month (DM 1),3 months (DM 3) and 6 months (DM 6) respectively ( n =12). The body weight and blood sugar were examined once a month. Then the retina were taken and were prepared as the ultrathin section and observed by transmission electron microscope. Results:The microvilli and infolds of plasmic membrane of retina pigment epithelium were a few and scattered,shortened and came off or lowered with the rise of blood sugar and the worsening of illness. The main impairment were swelling of mitochondrial,fall off of crista and enlargment of endoplasmic reticula,but the complex of junction between the pigment epithelium consistantly existed. Conclusion:The damage were mainly in the microvilli of plasmic membrane of basis of retina pigment epithelium in DR rats,but there were no damage to the choroid membrane retina barrier. This study provide the morphological evidences for the clinical research of the DR.
