摘    要:目的:建立以反相高效液相色谱法同时测定巴戟天中2-羟基-3-羟甲基蒽醌、2-羟基-1-甲氧基蒽醌、甲基异茜草素-1-甲醚、甲基异茜草素4种蒽醌类化合物含量的方法。方法:色谱柱为Extend-C1(8250mm×4.6mm,5μm),柱温为30℃,流动相为0.2%磷酸水溶液-乙腈(梯度洗脱),流速为1.0mL.min-1,检测波长为277nm。结果:2-羟基-3-羟甲基蒽醌、2-羟基-1-甲氧基蒽醌、甲基异茜草素-1-甲醚、甲基异茜草素进样量均在0.2~250μg·mL-1范围内与各自峰面积积分值呈良好线性关系(r=0.9999或0.9998);平均回收率分别为98.69%、99.67%、97.91%和99.74%,RSD分别为0.90%、1.97%、1.14%、0.41%(n=6)。结论:本法简便、准确、重复性好,可用于巴戟天中4种蒽醌类化合物的含量测定。

Abstract:OBJECTIVE:To study and establish the RP-HPLC method for the content determination of four kinds of anthraquinones from Morinda officinalis,such as 2-hydroxy-3-hydroxymethylanthraquinone,2-hydroxy-1-methoxy anthranquinone,rubiadin-1-methyl ether,rubiadin. METHODS:The separation was performed on Agilent Extend-C18(250 mm×4.6 mm,5 μm) column with mobile phase consisted of acetonitile-0.2% phosphoric acid (gradient elution). The column temperature was set at 30 ℃ and detection wavelength was 277 nm. The flow rates was set at 1.0 mL·min-1.RESULTS:The linear range of 2-hydroxy-3-hydroxymethylanthraquinone,2-hydroxy-1-methoxy anthranquinone,rubiadin-1-methyl ether,rubiadin were all within 0.2~250 μg·mL-1(r=0.999 9 or r=0.999 8). The average recovery rates were 98.69%,99.67%,97.91% and 99.74% and RSDs were 0.90%,1.97%,1.14%,0.41%(n=6),respectively. CONCLUSION:The method is simple,accurate and reproducible for the content determination of four kinds of anthraquinones from M. officinalis.
