白背叶黄酮类化合物抗鸭乙型肝炎病毒活性研究

摘    要:目的:研究白背叶黄酮类化合物(WF)抗鸭乙型肝炎病毒(DHBV)的活性。方法:对1d龄雏鸭人工感染DHBV,7d后成功复制鸭DHBV感染模型。分别灌胃75、150mg·kg-1的WF,或50mg·kg-1拉米夫定,每天2次,连续给药10d。斑点杂交法测定给药第5天、第10天及停药后第3天鸭血清中DHBV-DNA浓度。结果:75及150mg·kg-1WF均能显著降低模型鸭血清中DHBV-DNA浓度(P<0.01),且停药后反跳程度轻于拉米夫定。结论:WF有显著的抑制DHBV-DNA复制的作用,表明其可能有较好的抗乙型肝炎病毒的活性。

Abstract:OBJECTIVE: To study the activity of flavonoids WF extracted from Mallotus apelta against duck hepatitis B virus (DHBV). METHODS: One-day-old ducklings were infected with DHBV. 7 days later the DHBV-infectious model was establis- hed successfully. The model ducklings were administrated with WF of 75 and 150 mg·kg-1 or lamivudine of 50 mg·kg-1 b.i.d.via i. g. gtt for 10 days consecutively. Serums were collected at 5th and 10th day after drug administration and 3th days after drug withdrawal. The level of DHBV-DNA in serum was detected by dot-blotting hybridization test. RESULTS: 75 and 150 mg·kg-1 WF reduced the level of DHBV-DNA significantly(P〈0.01 ), moreover, the rebound effect after discontinuation of WF was lower than that after discontinuation of lamivudine.CONCLUSION: WF significantly inhibits DHBV-DNA replication in DHBV models, which indicates that WF offers sound effect against HBV.
