金标特异抗原单克隆抗体斑点免疫金染色用于检测脑囊虫病循环抗原的研究

应用金标记猪囊尾蚴囊液抗原为探针,以抗猪囊尾蚴抗原的McAb为联桥,建立了非常规Dot-IGS用于检测脑囊虫病患者CAg的方法,用4G_2McAb检测84例脑囊虫病人脑脊液(CSF),CAg阳性率为88.09％。检测63例其他疾病患者,出现2例假阳性。以1F_(11)McAb检测14例脑囊虫病人,13例检出CAg,阳性率为92.86％。检测其他疾病患者36例,均为阴性。用4G_2与1F_(11),株McAb可分别检出最小抗原量为1ng/ml和0.25ng/ml。本法敏感、特异、操作简单,可用于脑囊虫病的诊断。

STUDY ON CAg DETECTION OF PATIENTS WITH NEUROCYSTICERCOSIS BY SPECIFIC ANTIGEN-GOLD PROBE WITH McAb DOT-IGS

The Gold-labelled cyst fluid antigen of cysticercus cellulosae was used as a probe. The antigen-gold probe and CAg in CSF were linked by McAb against cyst fluid antigens of cysticercus cellulosae. Dot-IGS was established for detection of CAg in CSF of patients with neurocysticercosis. CSF of 84 cases with neurocysticercosis were detected with 4G2 McAb, the CAg positive rate was 88. 09% .13 of 14 cases with neurocysticercosis detected with 1F11 McAb were positive. 63 cases with other diseases were negative except two with 4G2 McAb. All 36 cases with other diseases were negative when 1F11 McAb was used. The minimal detectable level was 1 ng/ml(with 4G2 McAb) and 0. 25 ng/ml(with lF11 McAb) respectively. It suggests that this assay is sensitive,specific and simple and suitable for neurocysticercosis diagnosis.
